Suoquan Wan,a common prescription recorded in Fu Ren Liang Fang,consists of salt Alpiniae Oxyphyllae Fructus、Linderae Radix and Dioscoreae Rhizoma and is frequently used in curing enuresis and diuresis caused by Kidney-Yang deficiency.Salt-processing can inject drugs into kidney,and relieve its dryness.Salt processing makes Alpiniae Oxyphyllae Fructus,the Principal medicine of Suoquan Wan less acrid and dry,better at warming kidney、consolidating semen and reducing urine,and enhance efficacy of the whole prescription.Basing on previous studies,HPLC has been used in this thesis to establish and compare the fingerprint spectrum of Suoquan Wan(extracted with methyl alcohol and Petroleum ether)constituted with raw and salt-stiring Alpiniae Oxyphyllae Fructus;Grey correlation method has been used to analysis the relationship between different extract of Suoquan Wan and its urine-reducing efficacy;HPLC and LC-MS have been used to study its components absorbed into plasma and establish the fingerprint spectrum of plasma.These studies provide the basis for clarifying the material basis of enhancing curative effect after salt-processing of Alpinia.By comparing the fingerprint spectrum of the Suoquan Wan constituted with raw and salt-processed Alpinia,the No.1 peak of the relative peak area changed dramaticlly,and the increase rate is 183.33%;By studying the relationship between different extracts of Suoquan Wan and its urine-reducing efficacy,petrol ether extracted part of Suoquan Wan was confirmed to be the urine-reducing effective part,and the contribution of different chromatographic peaks on the efficacy of reducing urine was order as No.8>No.9>No.7>No.11>No.17>No.5>No.19 No.>No.29 >No.18 >No.10 No.>No.6 > No12.>No.23;By comparing fingerprint spectrum of effective part of Suo Quan Wan,the relative area peaks No.1,2,3,4,5,6,7,11,12,13,18 changed(change rate> 30%).No.1,No.4(teuhetenone A),No.5 and No.7 peak which have a great contribution to urine-reducing efficacy increase 123.08%,90.91%,53.33% and43.75%;Studying on the components of Suoquan Wan absorbed into plasma,7chemical components were found in plasma,including tectochrysin,chrysin,yakuchinone A,7-epi-teucrenone B,Alpinia oxyphylla,nootkatone and its metabolite.These components may be the substance basis of efficacy of Suoquan Wan.The plasma fingerprints of Suoquan Wan constituted with raw and salt processed Alpinia are similar,but the peak area changes.That means the effect may related to the change of the components’ contents.In this paper,fingerprint spectrum was used to reflect the changes of chemical composition out of and absorbed into plasma,and clarify the changes of substance basis after the salt-processing of Alpiniae Oxyphyllae.The spectrum-effect relationship and components into rat blood studies not only clarifies the mechanisms of therapeutic material basis,but also provides the basis of material base for the theory of salt processing of Alpiniae Oxyphyllae enhancing the kidney-warming and urine-reducing effect of Suoquan Wan.
