Fructus alpiniae oxyphyllae comes from the dry and ripe fruits of Alpinia oxyphylla Miq,which belongs to Zingiberaceae.According to the preliminary study of the research group,the effective component with “arresting polyuria” functions of the salt processed fructus alpiniae oxyphyllae is the petroleum ether.In addition,HPLC finger print is established for the effective component with “arresting polyuria” functions of the salt processed fructus alpiniae oxyphyllae.As the report of the present pertinent literature,the component with the pesticide effect of arresting polyuria is mainly in the essential oil or petroleum ether.Some other reports also say that it is mainly because of the sesquiterpenoids.Currently,there are no reports as to the piss shrink ingredients in the salt processed fructus alpiniae oxyphyllae.ObjectiveThis paper is aimed at clarifying the pharmacodynamic material basis of the salt processed fructus alpiniae oxyphyllae according to the chemical components represented by the characteristic peak on the finger print of the petroleum ether component of the salt processed fructus alpiniae oxyphyllae as well as its contribution to the inhibiting effect of the cavia procellus' s musculus detrusor vesicae.Moreover,it is going to discuss the effective ingredients of piss shrink in the petroleum ether component of the salt processed fructus alpiniae oxyphyllae.Furthermore,the pharmacodynamic verification is going to be conducted,intending to illustrate the pharmacodynamic material basis and action mechanism of piss shrink in the salt processed fructus alpiniae oxyphyllae.This paper focuses on studying the main active ingredients of the function change before and after the fructus alpiniae oxyphyllae being processed with salt,analyzes the comprehensive research thought and model employing by the single fructus alpiniae oxyphyllae medicine of “establishing spectrum-effect relationship-positioning active ingredients-the changes of single active ingredient”,reflects the dynamic change and transformation mechanism of the main active ingredients of the single fructus alpiniae oxyphyllae before and after the salt processing as well as explaining the processing theoretical mechanism of “being good at the kidney after salt process” and the basic theoretical connotation in the traditional Chinese medicine of “kidney controls the water”,thus accelerating the development of the theory of traditional Chinese medicine.MethodThis experiment optimizes the HPLC chromatographic condition,conducts segmentation research to the petroleum ether component of the salt processed fructus alpiniae oxyphyllae and employs gray correlation analytic approach to establish the spectrum-effect relationship of the petroleum ether component of the salt processed fructus alpiniae oxyphyllae,so as to discuss the piss shrink material basis of the salt processed fructus alpiniae oxyphyllae and further clarify its scientific connotation.In addition,the experiment separates and identifies the chemical ingredients of the petroleum ether component,so as to define its chemical structure.Moreover,leading by the medicine function,this experiment separates the medicine functions to the the petroleum ether component of the salt processed fructus alpiniae oxyphyllae and identifies several chemical compounds.After function verification,the chemical compounds have significant influences to the average tension and inhibition ratio to the shrink of cavia procellus' smusculus detrusor vesicae.Finally,the experiment initially reveals that the chemical compound7-epi-teucrenone in the salt processed fructus alpiniae oxyphyllae might be the effective substance of “warming kidney and shrinking piss”.ResultThe finger print of the petroleum ether component of the salt processed fructus alpiniae oxyphyllae has been established.The similarities are all above 0.9.The piss shrink function of the salt processed fructus alpiniae oxyphyllae is the common functions of several ingredients.The gray correlation analysis method is used to rank the sequence of the contributions of the chemical compounds represented by various characteristic peaks(according to the code of the characteristic common peaks).The sequence is of the salt processed fructus alpiniae oxyphyllae,nine chemical compounds have been identified:(nootkatone,11-hydroxy-valenc-1(10)-en-2-one,teucrenone,7-epi-teucrenone B,12-Hydroxynootkatone,tectochrysin,?-sitosterol,daucosterol,palmitic acid).After verification of pharmacodynamics,7-epi-teucrenone B has close relationship to kidney warming and piss shrinking.ConclusionThe established HPLC finger print of the petroleum ether component of the salt processed fructus alpiniae oxyphyllae has good repeat-ability,high stability and simple employing method.It can provide reference for the evaluation of the piss shrinking.The finger print of the petroleum ether component has quite distinct correspondence with the piss shrinking efficacy.Therefore,it lays foundation for the further study of the piss shrinking.The pharmacodynamic experiment is conducted to verify the influences of the separated chemical compounds of the petroleum ether component of the salt processed fructus alpiniae oxyphyllae to the cavia procellus' s musculus detrusor vesicae.The result indicates that 7-epi-teucrenone B might inhibit the shrink tension of the cavia procellus' s musculus detrusor vesicae through H1 receptor release of the antagonism of histamine phosphate.Therefore,it means that the 7-epi-teucrenone B in the petroleum ether component of the salt processed fructus alpiniae oxyphyllae is probably the main efficient ingredient of the piss shrinking function of the salt processed fructus alpiniae oxyphyllae.
