Experimental Study On The Effect Of Qi Fu Li Zhong Enema Decoction On PPAR γ/ NF-κB Signaling Pathway In Ulcerative Colitis Model Rats

Objective: Observed the pathological changes of colonic mucosa in rats with ulcerative colitis model and the effects of the expression of PPAR γ and NF-κB p65 in PPAR γ / NF-κB,and also the expression of ICAM-1 of Qifulizhong Enema Decoction to study the possible mechanism in the treatment of ulcerative colitis by using Qi fu li zhong Enema Decoction.Methods: 72 male SPF(specific pathogen free)grade Wistar rats were randomly divided into 6 groups: blank control group,model control group,sulfasalazine control group and Qifulizhong enema high,medium and low dose group that referred to as: blank group,model group,western medicine group,Qi-high group,Qi-medium group and Qi-low group.In addition to blank group,each group with TNBS/ anhydrous ethanol synthesis method of replication ulcerative colitis model,using 110 mg dose of TNBS each group(rat.)At the end of the modeling,the treatment was started on the 7th day.The western medicine group was enema with the sulfadiazine suspension and the enema dose was 315mg/d.The Qi-high group,Qi-medium group and Qi-low group were enema with Qifulizhong decoction,the dose ware 22.06g/d,11.03g/d and 5.52g/d.Observed the mental state,the activity of the rats and the other general events every day.In addition,measured the body weight,rectal temperature and fecal occult blood every 3 days at the end of the modeling and enema.After 14 days of continuous enema treatment,rats in each group were fasted for 24 hours,and anesthetized by intraperitoneal injection of 10% chloral hydrate.The femoral artery was taken for serum collection.Along the abdominal line to open the abdominal cavity and measured the distance from the anus 2-12 cm of intestinal section,then cut open.Naked eye observation of the colon tissue for injury score after PBS washed.Cut about 1cm the middle of the colon tissue and divided into two parts.Fixed with 4% paraformaldehyde solution to prepare the pathological,immunohistochemical and Western Blot detection.Result: 1.The mental status,feeding rate,stool condition,rectal temperature and body weight of the rats in each dose Qifulizhong Enema Decoction group were improved than those in the model group.2.Visual and microscopic observation show that hyperemia and edema of UC rats in each dose Qifulizhong Enema Decoction group was significantly lower than that in the model group.3.The results of immunohistochemistry: Compared with the blank group,the content of PPARγ in the colonic tissue of the model group was significantly decreased,the content of NF-κB p65 and ICAM-1 protein was significantly increased(P <0.05).Compared with the model group,the content of PPARγ in colonic tissue of Qi-high group,Qi-medium group and western medicine group was significantly higher than model group(P <0.05).The content of NF-κBp65 protein in Qi-high group and western medicine group was significantly lower(P <0.05.)The content of ICAM-1 protein in Qi-high group was significantly lower(P <0.05).The results of immunohistochemistry: 1.TNBS/ anhydrous ethanol method can successfully replicate ulcerative colitis rat model,and the duration of the disease is greater than 4 weeks.2.There is good therapeutic effect on the colonic mucosa in the UC model rats by using Qifulizhong Enema Decoction.The general condition and colonic pathology score of the rats were improved obviously.The mechanism may be related to up-regulation of PPAR γ expression in colonic tissue and inhibition of NF-κB p65 and ICAM-1 expression.
