Objective:To explore part of the mechanism of Pingkui Enema prescription on ulcerative colitis in rats.Methods:The first part of the experimental grouping normal group、model group、lzcl group、high、medium、and low joint(lzcl+pk of high、medium and low concentration)group.Lzcl groupgastric treatment;high、medium、low joint group with wypk enema treatment combined lzcl gastric treatment;The second part of the experimental grouping is normal groups、model group、lzcl group、low,medium,and high concentration groups of pk,all use enema treatment.The normal group and the model group were treated with distilled water enema treatmen.Each rat was fed 2ml·200g-1 for 14 day.ELISA detected levels of serum IL1-8 and IL-13;The content of bifidobacteria was quantitatively detected by rt-pcr.Results:The scores of DAI and HE staining in the model group were higher than those in the normal group(P<0.05),after medication,the DAI and HE scores decreased in each drug group(P<0.05).Compared with the normal group,the level of IL-8 in the model group was increased,which was statistically significant(P<0.05).Compared with model group,each treatment group rats IL-8 levels are lower,with low joint group effect is obvious,evey group of IL-8 levels compared with model group had statistical significance(P<0.05),.Compared with lzcl group,the IL-8 levels in all the joint groups were lower,and the comparison between the low joint groups and the lzcl group had statistically significant(P<0.05).Compared with the normal group,the level of IL-13 in the model group was decreased,and the comparison between them was statistically significant(P<0.05).Compared with the model group,the level of IL-13 in the joint group and the lzcl group were all increased,and the effect was significant with the the low joint group,and both were statistically significant(P<0.05).The levels of IL-13 in the rats of the joint group were all increased compared with that of lzcl.There was a statistically significant difference between the combination of the low joint group and the lzcl group(P<0.05).Compared with the normal group,the bifidobacteria mRNA in the model group was lower than that of the normal group.Group compared with model group,drug groups is higher than the model group,especially in the middle,the low concentration group significantly,compared with the Lizhu group,the low,medium concentration group is higher than the Lizhu,the high concentration group is lower than the Lizhu.Conclusion:Pingkui Enema prescription may reduce the level of IL-8 in serum and increase IL-13 in serum of UC rats,increase the number of bifidobacteria in the intestinal tract of rats with ulcerative colitis to achieve the goal of treating UC rats.
