| Purpose: Aim to observe the effects of sanbai enema liquid retention enema therapy onTNF-a in ulcerative colitis rats’ serum, using animal models to analyze the mechanism andstudy the advantages and therapeutic effects of sanbai enema liquid in treating ulcerativecolitis; in addition, to further reveal the superiority of traditional Chinese medicine retentionenema therapy for ulcerative colitis.Method:50healthy and clean Wistar rats are randomly devided into5groups, eachgroup rats are fasted but not inhibited from drinking water berfore modeling(except for blankgroup). The rats are anaesthetized with10%chloral hydrate (3ml/kg) and fixed on the ratdissecting table. Lubricate the polypropylene pipe (outside diameter2mm) with paraffin oil,and slowly insert into the colon,8cm of the rat anus’ upper segment. Then inject the5%TNBS and ethanol mix with the mixing ratio of12:5, each0.8ml(TNBS120mg/kg), lift therat tail and sustain inversion for1min, so the modeling agent can fully get into the ratintestine. Let the rat supine and tilt its head downward, pinch the anus2min until it naturallysober, then feed it normally. The sanbai enema liquid is divided into high and lowdoses(3.33g/kg,13.34g/kg) for treatment, positive group is treated with mesalamine(0.5g/kg),and set the model group and the blank control group. Record and score the general behaviorof the rats during treatment and on the14th day at the end of the treatment, observe and scorethe changes of the rats’ colon, then statistically analyze the results. Besides, taking about0.5cm pathological tissues for HE staining, and observe the histopathological changes ofcolon in each group under the optical microscope. At the end of the treatment, collect bloodserum samples and test the TNF-a level by ELISA, and statistically analyze the results.Result: The rats were decreased in the administration of24hours measuring bodyweight, with weight change after modeling, the rats in the model group the7d weight loss(P<0.05); the DAI score, the model group were higher than the other group (P <0.05), all themodel group DAI score was significantly higher than the control group, there was nosignificant difference between treatment groups. Observation of colon injury: in the modelgroup, single ulcer, ulcer found around the exudation, edema, three white enema in high dosegroup, a few inflammatory exudation, cells arranged in neat rows, low dose group of glands arranged irregularly, with inflammatory exudate, beautiful salad diazoxide group rats and theblank group similar. Serum TNF-a group rats were significantly higher than those in the blankgroup, there were significant difference between two groups (P<0.01). Three white enema inhigh dose group rats serum TNF-a levels lower than those in the model group, there was asignificant difference (p<0.01). The salad diazoxide group and three white enema high-dosegroup had no significant difference (P>0.05).Conclusion: This study found that, high-dose sanbai enema liquid group can reduceserum TNF-a levels of the rats, improve the local colonic mucosa damage and significantlyheal the ulcers, the study also shows that, sanbai enema liquid has a good therapeutic effectson ulcerative colitis rats. Modulating the immune, reducting colon inflammation, protectingintestinal mucosa and healing ulcer might be the curative mechanism of its effects onulcerative colitis. |
