Macleaya cordata(Willd.)R.Br.belongs to Papaveraceae,Macleaya R.Br.It has a lot effects such as sedative pain,anti-tumor,antibacterial and insecticide effect.It contains contains various chemical constituents such as alkaloids,flavonoids,terpenoids,steroidal and volatile oils.The main bioactive ingredient is isoquinoline alkaloids.In this study,the active tracer method was used to study the antibacterial active ingredients in the whole plant.It provided a theoretical guidance for the development of new plant-derived fungicide.The paper has obtained the following main results:1.Using ethanol ultrasonic extraction(solid-liquid ratio 10:1),the average extraction rate of the crude extract was 20.7%.2.The crude extract was dissolved in 25%aqueous methanol and the crude extracts were extracted with different solvents.The polarity was increased in order to obtain4 kinds of extracts:the petroleum ether phase(P),the chloroform phase(C),the n-butanol phase(B)and water-soluble substances(W),of which the highest content is petroleum ether phase,the lowest is n-butanol phase content.3.At 2 mg/mL concentration,the inhibitory activities of crude extracts and 4 extracts against 12 plant pathogens were tested.Tests have shown that the average inhibition rate of the chloroform phase and n-butanol phase is higher than that of the crude extract,the petroleum ether phase and the water-soluble substance.4.The gradient of chloroform and n-butanol phases was eluted by silica gel column chromatography.The elution gradient was chloroform:methanol=20:1,10:1,and 1:2(V/V).C1,C2 and C3 were gotten.The elution gradient of the n-butanol phase was chloroform:methanol=10:1,5:1 and 0:1(V/V),and B1,B2,and B3 were gotten.The above components were tested for antibacterial activity.The results showed that C1,C2 and B1,B2 have strong activity against plant pathogens.5.Seven kinds of monomer compounds were isolated from the active components(C1,C2 and B1,B2)by silica gel column chromatography and thin layer preparation.The structures were identified by chemical analysis,thin layer detection and NMR spectroscopy:BLH-1(ihydrosanguinarine),BLH-2(dihydrochelerythrine),BLH-3(sanguinarine),BLH-4(chelerythrine),BLH-5(6-methoxydihydrosanguinarine),BLH-6(protopine),BLH-7(allocryptopine).6.The activities of 7 compounds against 12 plant phytopathogen were tested.The results showed that sanguinarine,chelerythrine and 6-methoxydihydrosanguinarine which were separated from chloroform phase(C)have a broad-spectrum antimicrobial activity.7.The acidification n-butanol phase were extracted by n-butanol,we found that the organic components had higher activity than water phase.So we guess that the non-alkaloids ingredient was contributed to the major activity,it deserve a futher study. |
