High Altitude Polycythemia(HAPC)is called“Polyemia”.It is one of the most common chronic disease of serious threat to health of the plateau people.According to statistics,the prevalence rate of Polyemia is about 2.5%in the Qinghai Tibet plateau.Duoxuekang capsule is a experiential prescription which come from the famous Tibetan medicine master Troru Tsenam.It has definite clinical effect in Polyemia for many years.Its main effect is activating blood circulation to dissipate blood stasis and clearing away heat and toxic material.It is composed of Rhodiola crenulata,Phyllanthus emblica,Hippophae rhamnoides and Zingiber officinale.Studies have shown that Duoxuekang capsule can improve the body’s tolerance to hypoxia and resist fatigue.At present,there is no literature report about the extraction technology and composition analysis on Duoxuekang capsule.In addition,the emergence of the mixed use in the market situation from the rapid increase demand of Rhodiola.In this study,firstly,analyze the quality of Rhodiola crenulata(HK.f.et Thoms.)H.Ohba,Rhodiola kirilowii(Regel)Maxim and Rhodiola fastigiata(Hk.f.et Thoms.)S.H.Fu)by comparated its main chemical components and antioxidant activity.Then optimize extraction technology of Duoxuekang capsule by a uniform design method.Lastly,established the method of simultaneous determination of 5 kinds of chemical components in Duoxuekang capsule after identified the chemical components by Hyphenated technique,which can provide reference basis for the study of the material basis and quality control.Objective:1.Comparison of antioxidant capacity and chemical composition of three common Rhodiola plants.2.To optimize extraction technology of Duoxuekang capsule.3.To determine the composition and content of the chemical components in Duoxuekang capsule.Method:1.To establish an HPLC method for simultaneous determination of salidroside,gallic acid,tyrosol,caffeic acid,p-coumaric acid in R.crenulata,R.kirilowii and R.fastigiata.Combined with the in vitro antioxidant experiment,compare the differences of three common Rhodiola plants by statistical analysis.2.To optimize extraction technology of Duoxuekang capsule with salidroside,gallic acid and extraction yield as evaluation indexes by a uniform design method.3.To determine the composition of the chemical composition of Duoxuekang capsule with UPLC-Q-TOF-MS and GC-MS analysis4.To establish an HPLC method for simultaneous determination of salidroside,gallic acid,tyrosol,corilagin and ellagic acid in Duoxuekang capsule.Result:1.The content of salidroside,tyrosol,gallic acid,caffeic acid were in the order of R.crenulata>R.kirilowii>R.fastigiata.The content of p-coumaric acid were in order of R.kirilowii>R.crenulata>R.fastigiata.The DPPH·and ABTS·+radical scavenging ability were in the order of R.crenulata>R.kirilowii>R.fastigiata.The·OH radical scavenging ability were in the order of R.crenulata>R.fastigiata>R.kirilowii.2.The best content of extraction condition were achieved and listed as follows:the proportion of prescriptions weighed by medicinal materials,extracting with 50%alcohol,the proportion of solid to liquid was 1:14,infusing the soak time was 1.5 h,extracted 2 times with 1 h per time.3.LC-MS and GC-MS indicated that ethanol extract of Duoxuekang capsule mainly contains salidroside,gallic acid,crenulatin,Rhodalidin,corilagin,ellagic acid,chebulagic acid etc.The volatileoilmainlycontained1-octanol,zingiberene,sesquiphellandrene,linalool,(4Z)-4-(1,5-dimethyl-4-hexenylidene)-1-methyl-cyclohexene,curcumene etc.4.A good linear relationships of salidroside,gallic acid,tyrosol,corilagin and ellagic acid of Duoxuekang capsule were at 2.9423.023 mg·g-1,35.99636.044 mg·g-1,1.9722.006 mg·g-1,8.0098.085 mg·g-1,8.7298.820 mg·g-1,respectively.Conclusion:1.There were differences of antioxidant capacity and chemical composition in three common Rhodiola plants.R.crenulata is better than R.fastigiata and R.kirilowii.2.This research provided basis for extraction technology of Duoxuekang capsule.It was reasonable and simple of this extraction process.3.Established the method of simultaneous determination of 5 kinds of chemical components in Duoxuekang capsule was based on the analysis of chemical components,which can provide reference basis for the study of the material basis and quality control.
