The unique ecological environment of the Qinghai-Tibet Plateau gave birth to a wealth of Tibetan medicine resources,Living in the Tibetan Plateau Tibetan ethnic survival and reproduction after thousands of years,which has accumulated a wealth of experience in the use of plateau drugs and unique drug theory.Chinese medicine pay attention to the moral,the tibetan medicinal properties also have the impact of living environment,Both theories emphasize the quality of herbs closely related to their growing environment.Tibetan medicine mainly grows in the alpine meadow,alpine pad,mountain flow stone beach,its special growth environment restricts the Tibetan medicine cultivation and Tibetan medicine industry development.The main question now is how to combine the quality of medicinal herbs to guide the protection and cultivation of endangered Tibetan medicine under the guidance of the theory.In addition,due to geographical differences,Tibetan medicine for the local materials,the same kind of herbs,different areas of the use of base species are not the same and other issues lead to its mixed phenomenon is very serious.In addition,there are regional differences in Tibetan medicine,the same medicinal herbs,different areas of the use of base species are not the same and other issues,which result in its mixed phenomenon.Such as "Tida","Rhodiola" and other herbs have a variety of base,and multi-base mixed with the impact of the quality of Tibetan medicine and in-depth development and utilization,therefore,it is necessary to establish a analytical method for rapidly identify different original species,which ensure the safety of the base of the original medicine and legitimately use.In this study,taking "R.crenulata " as an example,we used the GIS technology and the ecological model to forecast the potential distribution area by investigating the status of its resources.The quality of the medicinal materials and the relationship between the ecological environment were analyzed by using UPLC-Q-TOF-MS,UPLC,FTIR and other technologies,which provide reference for the quality control of Tibetan medicine and the sustainable use of resources.Purposes:1.Investigation on the distribution and utilization of wild resources in rhodiola,to understand its resource reserves;collectting samples for rapid identification.Rapidly analysis the chemical composition of R.crenulata;a rapid method for the determination of 5 components(Gallic acid,Salidroside,Tyrosol,Ethyl gallate,p-Coumaric acid)in R.crenulata was established;The content of five components in Rhodiola in different regions was determined.2.Analysis on the suitability of the origin and quality regionalization of R.crenulata,Which guide artificial cultivation.Methods:1.Combined with literature survey,visiting investigation and field investigation,the quadrat was designed to calculate the amount of storage,and to record the information of habitat and latitude and longitude.Record its habitat,latitude and longitude and so on.2.according to the three grade identification model of FTIR,which included one-dimensional correlation IR(1D-IR)spectroscopy,second derivative IR(SD-IR)spectroscopy and two-dimensional correlation IR(2D-IR)spectroscopy,combined with stoichiometric methods can be identify and discriminate Rhodiola crenulata(Hook.f.et Thoms.)H.Ohba,R.kirilowii(Regel)Maxim and R.fastigiata(HK.f.et Thoms.)S.H.Fu.3.The chemical constituents of R.crenulata were identified by UPLC-Q-TOF/MS;The content of 5 compounds(Gallic acid,Salidroside,Tyrosol,Ethyl gallate,p-Coumaric acid)in R.crenulata was determined by UPLC-PDA.4.Predict suitable growth area and growth conditions of Rhodiola by Maxent niche model and geographic information system technology;The relational model between chemical components and ecological factors in Rhodiola was established by stepwise regression equation,then,the spatial distribution of chemical components of R.crenulata in different regions was predicted by GIS software.Results:1.R.crenulata grown in the Qinghai-Tibet Plateau elevation of 3200 ~ 5600 m area,Showing a flaky distribution,wild resources have been severely damaged,With the development of R.crenulata products,its demand is increasing.2.The correlation coefficient and partial least squares discriminant analysis based on the one dimensional infrared spectral data.which can be used to distinguish the three cultivars.3.Firstly,35 compounds were identified from the mass spectrometry of R.crenulata,by reviewing the literature and related databases including Gallic acid,Salidroside and so on.Secondly,A method for the determination of five components(GGallic acid,Salidroside,Tyrosol,Ethyl gallate,p-Coumaric acid)of R.crenulata was established,The method has the advantages of good separation effect,short analysis time,methodological study and sample recovery are in line with the requirements.The content of five components in 38 samples of Rhodiola was determined by this method.4.The results of Maxent model show that the potential distribution area of R.crenulata is in the eastern part of Tibet,western Sichuan,southern Qinghai,and Gansu Gannan Tibetan Autonomous Prefecture,Yunnan Diqing Tibetan Autonomous Prefecture.R.crenulata sachalinensis in some areas has higher content in R.crenulata,which is suitable for the cultivation of rhodiola.Including: Shiqu County,Litang County,Daocheng County in Sichuan Ganzi,Hongyuan County in Sichuan Aba Prefecture;Nagqu County in Tibetan Nagchu area,Milin County and long county in Tibetan Nyingchi area,Dinggye County and Lazi County in Tibetan Shigatse area,Gyaca Countyand Sangri County in Tibetan Shannan area;Nangqian County,Yushu County,and Maduo county in Qinghai;Deqin County,Shangri-La County in Yunnan and so on.ConclusionsAt present,there is a great demand for medicinal materials in R.crenulata,and the destruction of wild resources is serious.So we need to strengthen its wild resources protection and artificial cultivation to realize the sustainable development of its resources.According to the guiding theory of Tibetan medical,this study adopts the geographic information system and the ecological model,combined with the effective components for quality zoning analysis,Which can provide the scientific basis for the conservation and artificial cultivation of Rhodiola salsa,and also provide the methodological reference for the protection and sustainable utilization of other multi-base and endangered Tibetan medicines. |
