Objective: To explore the effects of overexpression of ALCAM in human hepatoma cells HepG2 cells in vitro and in vivo.Methods:1.Human hepatoma cell line HepG2 cells were infected with recombined adenovirus pSES-Ad-ALCAM to establish ALCAM overexpressed HepG2 recombined cell models.The expression of ALCAM protein in cells was detected by Western blot.2.CCK-8 and Flow cytometry were used to detect cell viability and apotheosis changes,respectively.Transwell was used to test the cell invasion and migration ability.3.Recombined adenovirus pSES-Ad-ALCAM and pSES-ALCAM-siRNA were injected into and around the tumor of nude mice to detect the effect of ALCAM on the growth of tumors.The expression of ALCAM protein and CD34 in tumors were tested by IHC.Results:1.Western blot results showed overexpressed ALCAM protein in Ad-ALCAM group compared with the control.2.The cell viability in the Ad-ALCAM group was increased tested by the CCK-8,the apoptosis rate of cells in the Ad-ALCAM group measured by FCM was decreased,and the cell migration and invasion ability in the Ad-ALCAM group had decline tested by Transwelll(P<0.05).3.In the mouse xenograft model,the tumor growth was accelerated in the fAd-ALCAM group,and the ALCAM protein and CD34 expressed was also increased compared with the control;the tumor growth was delined in the ALCAM-siRNA group and the ALCAM protein and CD34 expressed was decrease compared with the control group(P<0.05).Conclusion:ALCAM could increase the cell viability of Human hepatoma cell line HepG2,decreased the rate of cell apoptosis,and had an impact on cell migration and invasion.At the same time,ALCAM has been shown to promote tumor growth in vivo.
