| Objective :To detect the serum Galectin-3 level in population with different glucose tolerance and evaluate its correlation with pancreaticβ-cell function,insulin resistance,glycogen and lipid metabolism index.Methods: This study was conducted in Health Examination Center of Hebei Province General Hospital from January to April,2017.Eventually,a total of 268 individuals were enrolled in the study.All participants received75 g oral glucose tolerance test(OGTT)after signing the written informed consent form.According to the results of glucose tolerance test,the subjects were divided into three groups,including normal glucose tolerance(NGT),impaired glucose regulation(IGR)and type 2 diabetes mellitus(T2DM).Peripheral venous blood samples were collected from all participants.Serum was obtained by centrifugation at3,000 rpm for 10 min.after centrifugation,the upper serum was separated.The biochemical indexes such as blood glucose and blood lipid were detected by automatic biochemical analyzer.The level of serum insulin was determined by electrochemical luminescence method.The glycated hemoglobin(Hb A1c)was determined by high pressure liquid chromatography.Serum levels of Galectin-3,TNF-α(Tumor necrosis factor-α)and CRP(C-reactive protein)were detected by ELISA method.Homeostasis model assessment of insulin resistance(HOMA-IR)was used to evaluate IR,as calculated by the following : fasting blood glucose(FPG,mmol / L)x fasting insulin(FIns,m IU /L)/22.5.HOMA-β was used to assess pancreatic β-cell function,HOMA-β = 20 x fasting insulin(FINS)/(fasting blood glucose(FPG)-3.5)(%).ALL statistical analyses were performed using SPSS21.0 software.Results:1 Comparison of the general indicators of each groupThe general indicators of each group were analyzed.The results showed no significant difference among the three groups in terms of age,gender,BMI and WHR(all P> 0.05).The indexes of glucose and lipid metabolism in each observation group were compared.Compared with NGT group,the levels of FPG,PBG and Hb A1 c were significantly higher in IGR group and T2 DM group(P<0.01).The levels of FPG,PBG and Hb A1 c in T2 DM group were higher than IGR group(P <0.01).We observed the differences of HOMA-IR,HOMA-β among the three groups.HOMA-IR was gradually higher among the NGT group and IGR group,T2 DM group(P <0.01).HOMA-β was becoming gradually lower among the NGT group and IGR group,T2 DM group(P <0.01).Compared with the NGT group,the T2 DM and IGR group have higher TC and LDL-C,but TC and LDL-C only showed significant difference between NGT group and IGR group(P <0.01,P<0.05).TG was gradually higher among the NGT group and IGR group,T2 DM group(P <0.05).2 Comparison of serum galectin-3,CRP,TNF-α levels among NGT,IGR and T2 DM group.The results indicated that the serum levels of galectin-3,CRP and TNF-α in T2 DM group and IGR group were significantly higher than NGT group.Compared with the NGT group,The serum galectin-3 and TNF-αlevels were higher in IGR group(P <0.05,P <0.01).3 The correlation of serum galectin-3 with basic parameters,glycogen and lipid metabolism index.The results suggested that the serum galectin-3 in this study was positively correlated with FBG,PBG,Hb A1 c,TC,CRP,and negatively associated with HOMA-β.However,galectin-3 had no significant correlation with age,BMI,WHR,HOMA-IR,FINS,TG,LDL-C,TNF-α.In the study,we considered galectin-3 as the independent factor,using Multiple linear stepwise regression analysis to detected the influence of age,BMI,WHR,FBG,PBG,Hb A1 c,HOMA-IR,HOMA-β,FINS,TG,LDL-C,TC,CRP and TNF-α on the level of serum galectin-3.The result indicated that Hb A1 c was the important factor of serum galectin-3 level in people with different glucose tolerance.4 The association between serum galectin-3 level and the prevalence of type 2 diabetes mellitus.We divided all the participants into four groups across galectin-3quartiles(Q1:67.58-267.89pg/ml,Q2:267.90-415.92pg/ml,Q3:415.93-624.79pg/ml,Q4:624.80-1953.19pg/ml).The incidence of T2 DM with different galectin-3 levels were 23.88%,31.34%,40.30% and 58.21%,respectively,P<0.01.Compared with subjects in the first quartile,a significant increase of the incidence of T2 DM was observed in Q3 group and the highest quartile(P <0.05,P <0.01).Conclusions:1 Serum galectin-3 level was positively correlated with FBG,PBG,and Hb A1 c.2 Serum galectin-3 level was negatively correlated with HOMA-β.However,galectin-3 had no significant correlation with HOMA-IR. |
