Objective:To establish in rabbits with the experimental uveitis induced by bovine serum albumin and observe the change of symptoms and signs of rabbits’ eyes,and objectively evaluate the efficacy of Compound muyanning decoction as well as explore the expression levels of IL-2,TNF-α and ACA-IgG in the experimental uveitis.Methods:First,forty New Zealand white rabbits were randomly divided into four groups:normal control group,model control group,and TCM treatment group and hormone treatment group according to the random number table method.Second,bovine serum albumin(BSA)was used as an alloantigen to induce and establish the animal models in rabbits with experimental uveitis by secondary injection of vitreous and ear marginal veins.Third,0.25mL of 4%BSA was injected into the vitreous cavity of rabbits’s eyes;after one week,2.5mL of 2%BSA was injected into the ear vein of rabbits to establish an experimental animal model of uveitis.After the establishment of the model,the normal control group were not fed anything,and the model control group were fed with normal sal ine,and the control group of TCM was administered with Compound muyanning decoction as well as the hormone treatment group was given with the solution of prednisone acetate tablets.Last,the symptoms and signs of rabbits’ eyes in each group were observed by slit lamp microscope and ophthalmoscope,and the scores of ocular inflammation of rabbits were evaluated by Caspi criteria.At the same time,each rabbit’s aqueous humor and ear vein peripheral blood were collected at different time intervals(1st,3rd,7th,10th,14th,and 21st days),and the expression levels of IL-2,TNF-α and ACA-IgG in aqueous humor and peripheral blood of rabbits were detected at different stages by enzyme-linked immunosorbent assay(ELISA).Results:1 After modeling,the ocular inflammation scores of the model control group increased continuously.After administration,the ocular inflammation scores of the hormone treatment group and the TCM treatment group decreased at different degree respectiv-ely,and there was a statistically significant difference between two groups pre and post treatment(P<0.05).Further more,the ocular inflammation scores of the two treatment groups were significantly lower than the model control group(P<0.05),but there was no statistical significance between the two treatment groups(P>0.05).2 The expression levels of IL-2,TNF-α and ACA-IgG in peripheral blood and aqueous humor of rabbits in the normal group all had certain expression,but there was no statistical significance at different time points pre and post,and there was no statistically significant difference with other three groups before model in g(P>0.05).Af ter modeling,IL-2,TNF-α and ACA-IgG were gradually increased in aqueous humor and peripheral blood,and there was a statistically significant difference compared with the normal group(P<0.05).After medication,all indexes of the TCM treatment group was significantly lower than that of the model group at different time points(P<0.05).There was a statistically significant difference between pre and post treatment in the two treatment groups(P<0.05),and a similar effect to the hormone treatment group.Conclusion:1 The inflammatory response of rabbits with experimental uveitis induced by the bovine serum albumin can persist for more than 2 weeks.2 With the increase of the ocular inflammation scores in rabbits with experimental uveitis,the expression levels of IL-2 and TNF-α in serum and aqueous humor were also significantly increased,indicating that IL-2 and TNF-α are positively correlated with the development and progression of endogenous uveitis.3 Compound muyanning decoction can effectively control and improve the symptoms and signs of rabbit with experimental uveitis induced by BSA,reduce the ocular inflammation scores of the rabbits,and the levels of IL-2,TNF-α and ACA-IgG in serum and aqueous humor.In addition,the efficacy of compound muyanning decoction is similar to that of prednisone acetate tablets.4 Anticardiolipin antibody(ACA)-IgG is associated with the development and progression of endogenous uveitis.
