Objective:Establishment of rat model of threatened abortion with mifepristone,by observing the effects of flavored Andian Ertian Decoction on the general conditions of rats,the number of live births,the number of abortions and abortion rate,to evaluate the spuc curative effect of flavored Andian Ertian Decoction;by observing the changes of rat uterine decidual histomorphology,detection flavored Andian Ertian Decoction on the concentration of uterine decidual histomorphology TNF-α and IL-10,from Histomorphology and the imblance of T helper cells Th1(TNF-α)/Th2(IL-10)to discussion on the spuc mechanism of flavored Andian Ertian Decoction,to provide scientific experimental basis for clinical.Methods:(1)Grouping: 60 female SD rats and 30 male SD rats were selected,we put the estrous female rats and male rats at 2: 1 with the same cages overnight.the second day at 8 o’clock take female rats vaginal smear found a vaginal plug or sperm,It indicates that the female has been pregnant,on the first day of pregnancy,rats were randomly divided into 5 groups: The blank control Group,model group,progesterone group(western medicine control group),flavored Andian Ertian Decoction in high dose group and low dose group.(2)Dosing and modeling: from the 1-10 days of pregnancy,each rat of normal group was given 20ml/kg saline,from the 1-9 days of pregnancy,each rat of model group was given saline 20ml/kg;flavored Andian Ertian Decoction in high dose group and in low dose group each rat respective was given 33.48mg/kg,8.37mg/kg free fried granules,progesterone group at 1,3,5,7,9 days of pregnancy of each rat intramuscular injection of progesterone 6mg/kg.tenth days of pregnancy the rats in four groups(except normal group)were gavaged with mifepristone 4mg/kg,establish the model of threatened abortion.(3)Observation indexes and determination methods: five groups of rats were sacrificed on the twelfth day of pregnancy,shearing their uterus tissue,record bed points,record the bed points and the number of live births,calculate the number of abortion and abortion rate;separation of the left uterine decidua tissue,making slices,observe the histomorphology changes under the light microscope;take on the right side of uterine decidua tissue,using enzyme-linked immunoassay measured the concentration of IL-10 and TNF-a.Results:(1)The general condition of the rats: blank control group generally in good condition.model group after modeling general situation become worse.there is a small amount of bleeding seen from the vaginal opening.progesterone group,flavored Andian Ertian Decoction in high dose group and in low dose group,the three groups pregnancy rats after taking mifepristone generally have different degrees of variation.flavored Andian Ertian Decoction in high dose group is better,flavored Andian Ertian Decoction in low dose group and progesterone group variation degree are same,but they are better than model group.(2)Number of live births,number of abortion,abortion rate:model group’s number of live births lower than blank control group,number of abortion and abortion rate are higher than blank control group,there were extremely significant differences in statistical analysis(P < 0.01),progesterone group,flavored Andian Ertian Decoction in high dose group and in low dose group,the three groups’ s number of live births are higher than the model group,the three groups’ s number of abortion and abortion rate was lower than the model group,there were extremely significant differences in statistical analysis(P < 0.01);flavored Andian Ertian Decoction in high dose group and in low dose group,and progesterone group,comparison between the three groups,flavored Andian Ertian Decoction in high dose group’s number of live births higher than flavored Andian Ertian Decoction in low dose group and group progesterone,flavored Andian Ertian Decoction in high dose group’s number of abortion and abortion rate are lower than flavored Andian Ertian Decoction in low dose group and group progesterone,there were significant differences in statistical analysis(P < 0.05),flavored Andian Ertian Decoction in low dose group and group progesterone’s number of live births,number of abortion and abortion rate,the statistical analysis results showed no significant difference(P > 0.05).(3)the uterus decidua tissue of rats: blank control group’s organization structure is normal,morphology,distribution,and mass ratio between normal glands,no congestion,edema,diversification,differentiation of adenoid appearance change,the remaining muscularis is normal.model group’s most organization structure is destroyed,damaged area showed mild straight,Interstitial short spindle,more obese,rich blood vessels,involving the muscle;progesterone group,flavored Andian Ertian Decoction in high dosegroup,flavored Andian Ertian Decoction in low dose group have different degrees of damage.the extent of damage to flavored Andian Ertian Decoction in high dose group is lighter,flavored Andian Ertian Decoction in low dose group and progesterone group’s damage degree are same,but they are better than model group.(4)the concentration of IL-10 in uterine decidua tissue: blank control group’s the concentration of IL-10 is lower than blank control group’s,there is extremely significant differences in statistical analysis(P < 0.01).progesterone group,flavored Andian Ertian Decoction in high dose group and low dose group,the concentrations of IL-10 in the rat uterine decidua tissue of the three groups are higher than those in the model group,there is extremely significant differences in statistical analysis(P < 0.01).flavored Andian Ertian Decoction in high dose group and in low dose group,and progesterone group,comparison between the three groups,flavored Andian Ertian Decoction in high dose group’s the concentration of IL-10 is higher than flavored Andian Ertian Decoction in low dose group and progesterone group,there were significant differences in statistical analysis(P <0.05).Andian Ertian Decoction in low dose group’s the concentration of IL-10 compared with progesterone group,the statistical analysis results showed no significant difference(P > 0.05).(5)the concentration of TNF-α in uterine decidua tissue: model group’s the concentration of TNF-α is higher than blank control group’s,there is extremely significant differences in statistical analysis(P < 0.01).progesterone group,flavored Andian Ertian Decoction in high dose group and low dose group,the concentrations of TNF-α in the rat uterine decidua tissue of the three groups are lower than those in the model group,there is extremely significant differences in statistical analysis(P < 0.01).flavored Andian Ertian Decoction in high dose group and in low dose group,and progesterone group,comparison between the three groups,flavored Andian Ertian Decoction in high dose group’s the concentration of TNF-α islower than flavored Andian Ertian Decoction in low dose group and progesterone group,there were significant differences in statistical analysis(P <0.05).Andian Ertian Decoction in low dose group’s the concentration of TNF-α compared with progesterone group,the statistical analysis results showed no significant difference(P > 0.05).Conclusion:(1)Establishment of rat model of threatened abortion withmifepristone,flavored Andian Ertian Decoction in high dose group and in low dose can improve the general situation of rats,increase the number of live births,reduce the number of abortion and abortion rate,confirmed tocolysis effect of flavored Andian Ertian Decoction in high dose group and in low dose,and tocolysis effect of high doses is better than low dose.(2)flavored Andian Ertian Decoction in high dose group and in low dose can improve threatened abortion of rats’ uterine decidual histomorphology,it is conducive to embryonic implantation and development,high dose is better than low dose.(3)Andian Ertian Decoction in high dose group and in low dose can reduce the concentration of TNF-α and elevated the concentration of IL-10 in threatened abortion of rats’ uterine decidua tissue,makeing T helper cells Th1(TNF-α)/Th2(IL-10)type cytokines balance move to balance Th2,reduce the maternal-fetal immune rejection,high dose is better than low dose.
