factors.The abnormal accumulation of gene expression products and macromolecules,the activation of proto-oncogenes,and the suppression of tumor-suppressor genes are often involved in pathogenesis.Self-mutation of tumor biology is an important molecular event in the pathogenesis of gastric cancer.The study of molecular biology pathogenesis of gastric cancer is crucial for the diagnosis and treatment of gastric cancer and improve the prognosis of patients.With the in-depth research in the field of molecular biology,a large number of hot-spot molecules are continuously discovered and recognized.The PRDM14 gene is one of the hotspot molecules.Because of its unique gene structure and specificity of expression,it is considered that it may become a clinical diagnosis of gastric cancer and Effective targets for treatment.The PRDM family is a type of transcriptional regulatory factor that encodes a functional zinc finger protein,which mainly regulates gene expression and signal transduction,and participates in the physiological processes of cell growth,differentiation,apoptosis,etc.It is closely related to the pathogenesis of certain malignant tumors.The family member PRDM14 consists of one PR domain and six highly conserved zinc finger repeats.Expressed only in primordial germ cells and certain stem cells,the researchers screened for human genome-wide RNAi revealed that PRDM14 is a key transcription factor required to maintain stem cell characteristics and regain pluripotency in human somatic cells,and proves that it may be It is related to the onset of the tumor.At present,the progress of PRDM14 in cancer research has progressed.In breast cancer,PRDM14 has been shown to be associated with chemotherapy resistance and poor survival.Similar conclusions have been made in lung cancer research.Moreover,PRDM14 is also abnormally expressed in pancreatic cancer,HPV-positive tumors,and germ cell tumor tissues,and participates in the mechanisms involved in oncogenesis.PRDM14 may be a potential molecular target for cancer treatment.However,the expression and function of PRDM14 in gastric cancer is not clear.Objective To study the expression of PRDM14 gene in gastric cancer tissues,combined with basic data and clinical pathology factors of all patients included in the study,to analyze whether there is a correlation between different expressions of PRDM14 gene and clinical pathology factors of patients,and to investigate whether different expression of PRDM14 affects the prognosis of gastric cancer patients.Methods From January 2010 to September 2012,120 patients with gastric cancer who were admitted to the general surgery hospital of Zhengzhou University Affiliated Cancer Hospital were included in this study.All patients were obtained under gastroscopy or ultrasound gastroscopy before surgery,and pathology was identified as primary gastric cancer.Improve preoperative preparation by the same group of gastroenterologists for radical gastrectomy,surgical specimens within the specified time after the extraction of cancer tissue and adjacent normal gastric mucosal tissue,the specimens obtained by immunohistochemical detection of cancer The expression of PRDM14 protein in normal and normal tissues was collected.The basic data and detailed clinical pathology data of all patients were collected.The relationship between the expression of PRDM14 and clinical pathological factors in the enrolled patients was analyzed and analyzed.Long-term follow-up,statistical analysis of postoperative survival time,to investigate whether different expression of PRDM14 may affect postoperative survival of gastric cancer patients.Results 1.There was no clear PRDM14 protein expression in normal mucosa around the tumor.PRDM14 protein was positively expressed in 65(54.2%)patients in 120 tumor specimens,and negative expression of PRDM14 protein in the remaining 55(45.8%)patients;2.The expression of PRDM14 protein was related to tumor size,tumor location,depth of tumor invasion,lymph node metastasis status,lymph node staging,TNM stage(p<0.05),but not related to gender,age,tumor differentiation,vasculature and nerve invasion.(p all>0.05);3.Univariate Cox analysis showed that tumor size,lymph node metastasis status,lymph node staging,TNM stage and PRDM14 expression were the influencing factors of prognosis in this group of patients(p<0.05).Multivariate Cox survival analysis suggested that lymph node staging and PRDM14 expression Independent prognostic factors in this group of patients(p<0.05).Conclusion 1.PRDM14 negative expression in adjacent tissues,positive expression in some gastric cancer tissues,positive expression indicates poor prognosis;2.The expression of PRDM14 protein is related to tumor size,tumor location,depth of tumor invasion,lymph node metastasis status,lymph node staging,TNM staging,but not related to gender,age,tumor differentiation,vasculature and nerve invasion;3.The tumor size,lymph node metastasis status,lymph node staging,TNM stage,and PRDM14 expression are the influencing factors of the prognosis of this group of patients.Lymph node staging and PRDM14 expression as
