| Objective: The present study was designed to explore the relationship between HCRP1 gene expression and human gastric cancer,and combined with the clinical and pathological characteristics of the collected patients,we aimed to analyze the correlation between the expression of HCRP1 gene and clinicopathological features,and the effect of HCRP1 gene expression on prognosis of gastric cancer patients.Methods: The normal gastric tissues and gastric cancer tissues were collected from patients with gastric cancer,immunohistochemistry was performed to quantify the expressions of HCRP1 protein in gastric cancer tissues and non-cancerous tissues,and statistical method was used to analyze the results of immunohistochemistry and clinicopathological features.What’s more,we also used realtime quantitative PCR to quantify the expressions of HCRP1 mRNA in all cancer and normal tissues,and Kaplan-Meier survival analysis was used to analyze the relationship between the expression of HCRP1 protein and the prognosis of patients,COX regression model was used to analyze the prognostic factors of patients.Results: Immunohistochemistry results showed that HCRP1 is constitutively expressed in all GC tissues,and we found that the HCRP1 expression levels were significantly lower in cancer tissues compared with that in non-cancerous tissues(P<0.05).However,there was no significant difference between the expression of HCRP1 protein and gastric cancer tissues with different degree of differentiation(P>0.05).The expression of HCRP1 in GC tissues was closely correlated with tumor node metastasis(TNM)stage and distant metastasis(P<0.05).RT-PCR results showed that the expression of HCRP1 mRNA was highly expressed in normal tissues and low expressed in gastric cancer tissues and the difference was statistically significant.After the detection of RT-PCR,we also found that there was no significant difference between the expression of HCRP1 mRNA and gastric cancer tissues with different degree of differentiation.Kaplan–Meier analysis revealed that patients with high HCRP1 protein expression had a significant longer overall survival than patients with low HCRP1 expression(P<0.05).Univariate survival analysis showed that HCRP1 protein expression,lymph node metastasis,distant metastasis and TNM stage were prognostic factors,and Cox multivariate proportional hazards regression analysis showed that distant metastasis and TNM stage were independent prognostic factors.Conclusions : HCRP1 gene can inhibit the growth and increase of gastric cancer,and it was highly expressed in normal tissues and low expression in gastric cancer tissues.The expression of HCRP1 in GC tissues was closely associated with tumor node metastasis(TNM)stage and distant metastasis,and there was no association with patients’ age,sex,tumor size,and lymph node metastasis.It can be used as an index to predict postoperative survival in patients with gastric cancer,and it is also very important to evaluate the clinical stage and guide the treatment of gastric cancer. |
