| Background and purposeGastric cancer is a common upper gastrointestinal malignancy.The number of people with stomach cancer and the number of deaths in China account for about 1/2of the world.According to authoritative literature,in 2015,there were 679,000 new cases of gastric cancer in China,with a death rate of 498,000,ranking second in common malignant tumors and third in death.About 80%of them have been locally advanced gastric cancer at the time of initial diagnosis.At present,the treatment mode combined with adjuvant chemotherapy and radiotherapy is used.Although the chance of radical treatment is greatly improved,the 5-year survival rate is still low.Therefore,understanding the molecular biological changes in the development of gastric cancer,in-depth study of the pathogenesis of gastric cancer,and exploring new targets for the treatment of gastric cancer is an important aspect to improve the therapeutic effect of gastric cancer.PRDM14 is a member of the zinc finger protein family of positive regulatory regions and is an important molecule regulating epigenetic expression of genes,which plays a role in primordial germ cell specialization,cancer cell metastasis,invasion and drug resistance.Previous studies have found that PRDM14 is associated with breast cancer chemotherapy resistance and liver metastasis of pancreatic cancer,and similar findings have been found in bladder cancer and other cancers.However,the expression of PRDM14 in gastric cancer and its relationship with the development of gastric cancer have not been reported.This study intends to understand the expression of PRDM14 in gastric cancer tissues by immunohistochemistry,and to understand its effect on the migration ability of gastric cancer cells by lentiviral knockdown.Objective and methods1.To review the clinical data of patients who were diagnosed as gastric adenocarcinoma underwent radical surgery from December 2016 to February 2017 in the Department of General Surgery,Zhengzhou University,and randomly selected 40patients without anti-tumor treatment and complete medical history data.Inclusion in the study.Immunohistochemical staining was used to understand the expression of PRDM14 in the specimens and adjacent tissues of the patients,and the relationship between the expression and clinical pathology was analyzed by SPSS statistical software.2.The PRMM14 stable knockdown cells were constructed by lentiviral transfection in gastric cancer cell lines,and the relationship between PRDM14 and gastric cancer cell metastasis and EMT was studied by Western blot,scratch assay and Transwell migration assay.ResultsA total of 40 patients were enrolled.Immunohistochemical staining showed that PRDM14 could be expressed in the nucleus of gastric cancer.The immunohistochemical score of cancer tissue was significantly higher than that of adjacent tissues(6.925±2.443 vs 5.350±2.213,p=0.005).In the cancer tissues,PRDM14 was highly expressed in 22 cases(55%)and low in 18 cases(45%).Tumor lymph node metastasis was associated with PRDM14 expression(χ~2==8.741,p=0.047),and the difference was statistically significant(p<0.05).Patient gender(χ~2=0.404,p=0.525),age(χ~2=0.269,p=0.604),tumor site(χ~2=0.380,p=0.904),tumor size(χ~2=0.404,p=0.525),lauren classification(χ~2=5.618,p=0.060),degree of differentiation,(χ~2=1.410,p=0.726),T stage(χ~2=2.429,p=0.324),TNM stage(χ~2=3.160,p=0.175),vascular tumor thrombus(p=0.492),and neurological invasion(p=1)were not associated with PRDM14 expression,and the differences were not statistically significant.After knocking down PRDM14,the scratch test and Transwell migration test showed that the migration ability of gastric cancer cells decreased.After knocking down PRDM14,Western blot method was used to detect EMT-related proteins:E-cadherin expression was increased,N-cadherin expression was decreased,suggesting gastric cancer cells It is possible that EMT affects gastric cancer cell migration.Conclusion1.PRDM14 expression in gastric cancer tissues is higher than cancer side.Its expression is related to lymph node metastasis of gastric cancer;it has nothing to do with gender,age,tumor location,tumor size,lauren classification,differentiation degree,depth of invasion,TNM stage,vascular tumor thrombus and nerve invasion.2.Knockdown of gastric cancer cell PRDM14 can reduce the migration ability of gastric cancer cells. |
