Establishment Of Stem Apex Detoxification And Fast Propagation System On Two Strawberry Varieties

In recent years,facility agriculture in Inner Mongolia was vigorously promoted and strawberry was expanding rapidly because of its short growth cycle and high economic value.Especially in Hohhot,Baotou and Erdos area,the number of strawberry picking gardens has increased rapidly.However,the rapid development of strawberry planting in Inner Mongolia area were not balance with its strawberry seedling market shortage,which restricted the overall development of strawberry in Inner Mongolia.The present study showed that the establishment of the strawberry tissue culture system had a significant effect on the development of local strawberry seedling market.‘Red cheek’ and ‘asu’ strawberry were selected as tested materials and the fast propagation systems of stem tip detoxification of two strawberry varieties were established through the basic process of induction of shoot tip,axillary bud proliferation,rooting culture,transplant and domestication.The results would provide a theoretical basis for development of virus-free strawberry seedlings in greenhouse in Inner Mongolia.The results were as follows:1.The hormone ratio of 0.5mg/L 6-BA + 0.1mg/L IBA promoted the germination of ‘red cheek’ strawberry shoot tip and the germination rate reached 83.3%.In the medium of 0.5mg/L 6-BA + 0.2mg/L IBA,the germination rate of ‘asu’ strawberry reached the highest level of 75.0%.2.The 0.5mg/L 6-BA + 0.1mg/L IBA medium was the best of the hormone combinations in ’red cheek’ strawberry.‘Asu’ strawberry had the highest proliferation coefficient in the medium of 0.5mg/L 6-BA + 0.2mg/L IBA.The plant height of ’red cheek’ strawberry in the 0.5mg/L 6-BA + 0.1mg/L IBA medium was 7.7cm;and the plant height of ‘asu’ strawberry in 0.5mg/L 6-BA + 0.2mg/L IBA medium was 7.5cm.3.Rooting culture of ‘red cheek’ strawberry and ‘asu’ strawberry in 1/2MS medium and 0.1mg/L IBA were the best.The rooting number and root growth ratio reached the highest.4.With 0.3mm length of stem tips of the two strawberry varieties,the ‘red cheek’ and ‘asu’ strawberries were completely detoxified.5.Seed germination box was used for seedling cultivation.The survival rate of seedlings was higher than that of traditional seedlings...
