Study On The Rapid Propagation Of Yishui Lily By Tissue Culture

Lilium.spp is a perennial flower plant of Liliaceae and Lilium.It has high ornamental value,edible and medicinal value.Its bulb can be used to treat various diseases such as lung disease and anemia,which is the main ingredient of many tonics.Modern lilies are mainly planted,but lily has a long growth cycle and is highly dependent on natural conditions and external factors.In recent years,a large number of farmers have planted lily for poverty alleviation.Due to the lack of farmers' knowledge and technical constraints,the development of the entire lily industry has been seriously hindered,which is not conducive to the preservation of lily germplasm resources.The experiment was carried out from Lishui Lily in Shandong Province as an experimental material.The well-developed lily stems were selected as explants,and an efficient regeneration system was established for lily by tissue culture technique.In this experiment,the effects of different concentrations of 6-BA and NAA on the primary culture of lily,and the effects of different types and concentrations of auxin and cytokinin on the subculture of lily were studied.Moreover,the effects of different concentrations of medium and auxin on the rooting culture of lily were studied.Finally,an efficient regeneration system of lily was established.The results of this study are as follows:1.Primary cultureThe most suitable medium for the primary culture of Lily water lily is MS+1.0mg/L6-BA+0.1mg/LNAA.The value-added coefficient is 6.90,and the number of adventitious buds is 207.2.SubcultureThe most suitable cytokinin for subculture of Lilium sinensis is CPPU.When the CPPU concentration is 1.0 mg/L,the value-added coefficient is 8.40,and the number of adventitious buds is 252.The most suitable medium for subculture of Lilium sinensis is MS+1.0mg/LCPPU+0.05mg/LNAA.3.Rooting cultureThe most suitable auxin species for rooting culture of Lilium sinensis is NAA,and the most suitable concentration is 0.1 mg/L.The most suitable medium for lily root culture is 1/2MS+0.1mg/LNAA,the rooting rate reached 96.67%,and the average rooting number is 8.7.
