Jute(Corchorus spp.),including white jute(C.capsularis)and dark jute(C.olitorius),is one of the most important natural fiber crops,which is second to cotton in production area and yield.The new variety protection regulations must have the characteristies of DUS(distinctness,uniformity and stability)and proper names.Theguidelineof DUS testing is a technical document and also is a known method for new variety registry.Due to the late start of the development of guidelinefor jute DUS testing,there is no corresponding DUS testing guideline for jute at present.From this point,screening of example varieties andpolymorphism analysis of molecular markers as well as statistics analysis on morphological characteristics ofthe example varieties injute DUS testing were eonducted in this study,the main results are as follows.1.To screen example varieties in jute,a primary collection of 54 pre-selected example varieties was sampled on the basis of a core germplasm derived from 11 countries and regions.Diversity analysis were performed by the software of PopGen 32 and cluster analysis of these accesions by the software of NTSYS were investigated on the basis of 46 morphylogical characteristics at the farm of Yangzhong Science and Teaching of Fujian Agriculture and Forestry University in 2016 and 2017.The results showed that a total of 212 and 256 alleles for 46 morphological characteristics were observed in the white(34 accessions)and dark(20 accessions)jute respectively,with an average of 3.4068 and 4.4407 alleles for eachcharacteristics in the two cultivated species.The Shannon's diversity index averaged from 46 morphological characteristicswere 0.8318 with a range of 0 to 1.7674 in white jute and 0.9828 with a range of 0-1.8547 in dark jute respectively.These results indicated that the 54pre?selected example varietieswith rich diversity in morphological characteristics could be used for demonstration and adjusting for characteristics'describing.A collection consisted of 51 accessions will be used in the jute DUS testing after eliminating genetic redundancy of similar varieties.2.Studies on assessment of 46 morphological characteristics were conducted on the basis of screening of example varieties.Data analysis of these characteristics in two years indicated that 23 quality or psedo-quality characteristics basically were unchanged and 15 qualitative characteristics less affected by environments.These 38 characteristics eould be taken as manotory investigating characteries for each variety testing.Among them,the characterics of silique shape,axillary bud,location of flower and fruit,color of stem,color of petiole are grouping ones.3.Considering of construction of the fingerprint of jute DUS example varieties will facilitate the identification and protection of jute varieties,a total of 33 allelses were generated from 26 SSR or InDel primers with 51 example varieties.The polymorphic information content per molecular marker ranged from 0.1136 to 0.7512.The 51 jute varieties could be distinguished uniquely by these markers.The fingerprint of each variety will provide technoligical supports for conservation and registry of jute varieties.This study not only provides theoretical basis for selecting example varieties for jute DUS testing from different jute germplasm resources,but also gives a basis for determining testingcharacteristics,which will facilitate the development guidelines for the conduction of DUS testing in jute.
