Vitamin B is one of the essential organic compounds of human beings,and it plays a crucial role in the body's normal physiological metabolism.They have active properties,and are relatively stable under alkaline conditions,but is extremely unstable in heat,light conditions,and its structure would be susceptible damaged by oxygen,water,ultraviolet ray and many other external factors greatly when exposed to air,which greatly limit the application of vitamin B substances on the market.Using microencapsulation,vitamin B was embedded in the stable wall materials,which can effectively ensure the inherent properties of vitamin B,and improve its stability and oxidation resistance,which can reduce the loss of vitamin B products in its transportation,storage and other processes and decrease the damage of the environment outside of it.This experiment used KGM which is cheap and rich in resources,and gelatin as wall material substances,prepared micro capsules of vitamin B by using the mixed hybrid and freeze drying technique,measured and analyzed a range of indicators of vitamin B microcapsules.The main contents and conclusions are as follows:(1)Studied some properties of KGM and gelatin,which were vitamin B microcapsule wall material substances,made membrane by compounding the two materials,researched and analyzed the effects of several parameters(the ratio complex gum solution concentration and pH of the two wall material substance)on the complex apparent viscosity,the tensile strength and breathability of the film made by the mixed gel solution,and then obtained the by optimum conditions by response surface optimization,which were:concentration 2.13%,KGM/Gel 1.46,pH 8.13,and the tensile strength 42.9 MPa.(2)By usinng KGM and gelatin as wall material,thiamine hydrochloride(VB1),riboflavin(VB2),pyridoxine hydrochloride(VB6),folic acid,and a mixture of nicotinamide as the core material,adopting freeze-drying process,this experiment prepared water-soluble compound vitamin microcapsules.Researched and analyzed the effects of several parameters(wall material concentration,KGM/gelatin ratio,ratio of wall material core material)on the microencapsulation rate and capacity value,and then optimize the optimum conditions by response surface,which were:wall material concentration 2.13%,KGM/Gel ratio 1.32,wall material/core ratio 10.67,and under this condition the microencapsulation was 78.82%.(3)After the evaluation of physicochemical properties of the preparation of freeze-dried microencapsulated vitamin B product,the results showed that:the outlook of the product was more smooth,uniformed particles,compacted structure,and no other peculiar smell.Moisture content 3.67%,payload 56.46%,bulk density 0.47g/cm3,angle of repose 41°,which meant that it had a better description of the flow properties of the microcapsules.Used the scanning electron microscopy to observe the surface structure of vitamin B microcapsules products,and the results showed that the complex morphology of the microcapsules product was more beautiful and complete when compared with the single KGM and gelatin microcapsule.It combined the advantages of the two materials,and microcapsules were a little roll forming and were flat pieces.The infrared spectroscopy anylyzed complex microcapsules and draw the conclusion that it did not produce or disappear new or older perssads,but just enhance or offset the specific peaks,with no other changes.Through test results of the light,heat,storage stability at room temperature,vitamin B complex microcapsules,when contrast with single KGM and gelatin microcapsules,under the same conditions,had the highest content of vitamin B2 complex preservation rate,which illustrated that the stability of the vitamin B complex microcapsules was best.(4)In order to judge whether the vitamin B microcapsule prepared by experiments would cause hazards to the health of human beings,this research take tests of safety of the toxicological.Through the acute toxicity median lethal dose(LD50)test,observed the mouse had normal life,nand did not appear death or any other peticular negative phenomena.The mouse bone marrow cells micronucleus test and sperm aberration test showed that mouse micronucleus test and sperm abnormality changes are within permissible range,and did not occur significant differences,which indicated that the microencapsulated vitamin B did not induce variability and sperm aberration on the test mouse micronucleus.And the effects of 30d fed different doses of vitamin B microcapsules on subjects rat growth,blood biochemical parameters,reproductive system,organ functions were not significantly.Consolidated statement showed that the vitamin B microcapsule prepared by experiments are non-toxic and harmless,which is safe and reliable in the clinical use.
