Objective:Codonopsis pilosula is a traditional Chinese medicine for tonifying deficiency and invigorating spleen.At present,the production conditions of Chinese herbal medicine slice production enterprises are uneven,crude material,decoction pieces and rice fried pieces of Codonopsis pilosula are lack of unified quality standard system and processing technology standard,which limits the industry development of Codonopsis pilosula.The assay methodology color lustre,chemical index and HPLC fingerprint was studied,and the evaluation system of multi-index quality of Codonopsis pilosula was established,and the basic research on the standardization of processing technology among crude material,decoction pieces and rice fried pieces of Codonopsis pilosula were carried out.The changes of main chemical index and color lustre before and after processing of Codonopsis pilosula were analyzed,and the changes of material base and quality of Codonopsis pilosula were studied by comparing the HPLC fingerprint before and after the processing of Codonopsis pilosula,to provide data and theoretical support for standardization of TCM.Methods:(1)Establish the assay methodology of polysaccharide and Lobetyolin by HPLC,HPLC method was used to determine the content of Lobetyolin and 5-HMF,and Phenol-sulfuric acid method combined with UV was used to determine the assay methodology of polysaccharide.The method of colorimetric determination of Codonopsis pilosula was studied,and the Methodology Research on HPLC fingerprint of Codonopsis pilosula was established and studied.(2)The single factor experiment was carried out with the content of Ethanol extractives,polysaccharide and Lobetyolin as the index,and the data were analyzed by the Normalized distance evaluation method.The soaking time,moistening time,drying temperature,drying time and section thickness of Codonopsis pilosula were selected.(3)In this study,the color brightness(L*),red-green chromaticity coordinate(a*),yellow-blue chromaticity coordinate(b*)and total color value(E*ab)were used as the shape index.Ethanol extractives,polysaccharide,Lobetyolin and 5-hydroxymethyl furfural were used as Chemical index.The two index were adopted to optimize processing technology of rice fried Radix Codonopsis by taking processing temperature,processing time,the types and dosage of rice as factors.Normalized distance evaluation method is also used to optimize the experimental data.The processing technology of Rice fried Radix Codonopsis was optimized by orthogonal selection,and the correlation between the two indexes was analyzed by SAS 9.40 and JMP11.0.(4)Eleven batches of Codonopsis pilosula from similar sources were collected and processed according to the processing technology of the selected Codonopsis pilosula and rice fried Radix Codonopsis.The chemical indexes and color lustre among crude material,decoction pieces and rice fried pieces of Codonopsis pilosula,were analyzed.Combined with the fingerprints of Codonopsis pilosula and its processed products,the material basis of Codonopsis pilosula was compared before and after processing.Results:The methodology of determining the content of index components was good,and the method could be used to evaluate the quality of Codonopsis pilosula and its processed products,and the HPLC fingerprint of Codonopsis pilosula was precise,stable and reproducible.It can provide reference for qualitative identification of Codonopsis pilosula and its processed products.(2)The best preparation technology of Codonopsis pilosula is:Washing Codonopsis medicinal materials with the flow of drinking water quickly,place them in a covered enamel plate,add water until they have not exceeded the medicinal materials,soak for 1 hour,put the medicinal materials in a moisturizing pool after soaking,add water for 1:1,suffocate the medicine for 6 hours,adjust the distance between cutting knives,Slice thickness is 2~4 mm,dry at 60 ℃ for 10 h,remove debris after screening,set aside.(3)The cooking temperature,cooking time,amount of rice and type of rice have significant effects on the processing process of rice fried Codonopsis pilosula,and there is a significant positive correlation between the determination of chemical index and the result of shape index,and the processing technology of Codonopsis pilosula is to use japonica rice as raw material.When the stir-fry machine reaches 140 ℃,pour the slices and rice into the stir-frying machine,stir fry 10min.(4)There were significant differences in the chemical composition of crude material,decoction pieces and rice fried pieces of Codonopsis pilosula.There were 13 peaks in 11 batches of Codonopsis and Codonopsis pilosula and 14 peaks in rice fried Codonopsis pilosula.The similarity between different batches of crude material,decoction pieces was higher,most of which was greater than 0.9,and the similarity between batches of rice fried Codonopsis pilosula was poor.Conclusion:The index content determination method and HPLC fingerprint of Codonopsis pilosula can be used as a reference method for the quality study of Codonopsis pilosula.The processing technology of the selected decoction pieces and rice fried pieces of Codonopsis pilosula provides a way of thinking for the standardization study of traditionalChinese medicine.The changes of chemical composition and fingerprint before and after the preparation of more than 10 batches of Codonopsis pilosula are provided.It provides scientific basis for improving the quality evaluation standard of Codonopsis pilosula.
