Objective: Taking the commercially available Achyranthes bidentata and Codonopsis pilosula as the research objects,conduct systematic research on the identification,inspection,content determination,fingerprints and other aspects of their raw and processed products,compare the quality differences between the two,and comprehensively evaluate the commercial Achyranthes bidentata and Codonopsis pilosula The quality of decoction pieces provides an experimental basis for perfecting the quality evaluation system of two kinds of traditional Chinese medicine decoction pieces.Methods:Refer to the 2020 Chinese Pharmacopoeia to identify Achyranthes bidentata,Codonopsis pilosula and processed products by thin-layer chromatography;check its moisture,total ash,and extract;determine the content of its major components by ultraviolet spectrophotometry;determine by high performance liquid chromatography Its main component content;use high performance liquid chromatography to establish fingerprints and use "Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System(2012 Edition)" for similarity evaluation,determine their common peaks and perform cluster analysis;The phenol-sulfuric acid method was used to determine the polysaccharide content in raw products of Achyranthes bidentata and Codonopsis pilosula,use PMP pre-column derivatization Method to analyze the polysaccharide index components of Achyranthes bidentata and Codonopsis pilosula.Results: 1)TLC identification results showed that the two kinds of decoction pieces showed spots of the same color on the corresponding position of the reference substance.2)The moisture,total ash and extract content of 10 batches of Achyranthes bidentata decoction pieces and wine roasted products meet the requirements of the 2020 edition of the Pharmacopoeia,and the total sterone content before and after processing is in the range of 0.34%~0.56% and0.37%~0.62% respectively;The content of saponin was in the range of 1.36~2.20%,1.41%~2.44%;the content of β-ecdysterone was in the range of 0.069%~0.086%,0.061%~0.080%.The difference in total saponins content before and after processing is statistically significant(P < 0.05),indicating that total saponins can be used as an index component to distinguish raw products from processed products;the similarity of fingerprints of raw Achyranthes bidentata decoction pieces and wine and broiled products are both greater than 0.90,The composition is reduced after concoction.The results of cluster analysis before and after processing were inconsistent,indicating that the processing process would affect the chemistry of Achyranthes bidentata decoction pieces.Ingredients and content.Through the analysis of the monosaccharide fingerprints,five monosaccharide components were determined.The similarity of the fingerprints of the established achyranthes bidentata polysaccharide was greater than 0.90.3)The content of moisture,total ash and extract(except S6-1 and S9-1)of 10 batches of raw dangshen decoction pieces and rice stir-fried products are in compliance with the regulations of the2020 edition of the Pharmacopoeia,and the total saponins content before and after processing are respectively 0.90%~1.39%(S4 Except),within the range of 0.75%~1.07%;the total flavonoid content is within the range of 0.28%~0.36%,0.24%~0.81%;the content of Codonopsis is within the range of 0.018%~0.071%,0.016%~0.050%.The differences in moisture,ash,total flavonoids and codonopsis before and after processing are statistically significant(P < 0.05),indicating that these four components can be used as index components for distinguishing raw and processed products;fingerprints of Codonopsis and rice fried Codonopsis greater than 0.90,the number of peaks increased after processing.The results of cluster analysis before and after processing are inconsistent,indicating that the processing process will affect the chemical composition and content of Dangshen decoction pieces.Three monosaccharide components were determined by analyzing the fingerprints of monosaccharides,and the similarity of the fingerprints of the polysaccharides of Codonopsis pilosula was greater than 0.90.Conclusion: The method for determining the content of index components in the prepared decoction pieces and the HPLC fingerprint spectrum are stable,reliable,and characteristic.They reflect the changes in the composition of the decoction pieces of traditional Chinese medicine before and after processing,and basically reflect the overall chemical composition characteristics of the decoction pieces of Achyranthes bidentata and Codonopsis pilosula.Its quality control provides the basis.
