Purpose : To observe the effects of serum containing Zuogui pill substances on the expression of GPR48, BMP7, BSP, ATF4 and TGF-β in rat bone marrow mesenchymal stem cells (BMSCs) in vitro cell culture, based on the TCM theory of "The kidney generating marrow and dominating bone". To explore the mechanisms of efficiency of the prevention and treatment of osteoporosis with Zuogui pill, so as to provide experimental basis for the theory of "The kidney generating marrow and dominating bone".Material and method: Forty female rats aged 3 months with no mating of SPF grade were used in this study, which weighted 230-250g. Forty rats were randomly divided into normal group, induced liquid group, Zuogui Pill group, Fosamax group and GSK group. The corresponding drugs were given for 5 days and the serum of each group was collected. The bone marrow mesenchymal stem cells were cultured in each group. The results of differentiation of rat bone marrow mesenchymal stem cells into osteoblasts were observed.The protein levels of ALP, GPR48, ATF4, BSP, BMP7 and TGF-P were measured by ELISA.The SPSS17.0 statistical software were used to analyze the result of serum containing Zuogui Pill substances.Results:1. The results of MTT in each experimental group were shown as follows: Compared with the normal group, the Zuogui pill group was significantly higher than that in the normal group (P<0.01). Compared with the Induced fluid group, Fosamax group and GSK group, there was significantly different (P <0.01).2. ALP results in each experimental group: There was significant difference between the Zuogui pill group and normal group (P <0.01). There was significant difference between the Zuogui pill group and Fosamax group, GSK group (P <0.01).3. The protein contents of GPR48, ATF4, BSP, BMP7 and TGF-β in rat bone marrow mesenchymal stem cells were determined by the serum of each group: Compared with the normal group, the other four groups were significantly improved (P <0.01).Conclusion:1. GPR48, BMP7, BSP, ATF4 and TGF-β could be expressed in normal rat serum, indicating that GPR48, BSP, ATF4, BMP7 and TGF-β signal factors were present in rats.2. Serum Containing Zuogui Pill Substances can improve the serum levels of GPR48, BMP7,BSP, ATF4 and TGF-β, indicating that it may regulate the protein content expression of GPR48, BSP, ATF4 and TGF-β to achieve the purpose of prevention and treatment of osteoporosis.3. As a representative of traditional Chinese medicine kidney remedies, Zuogui Pill can promote bone marrow mesenchymal stem cells to osteoblast differentiation, to a certain extent,verify the scientific theory of "kidney governs bone and marrow".
