Purpose:In this study,the female ovaries were removed to model the postmenopausal osteoporosis animals.The rats were treated with Bushenjingjing Zhuanggu Recipe Zuogui Pills to observe the changes of bone mineral density,body weight and uterus wet weight.To detect the changes of SOD,NO,GSH-Px and MDA levels in the serum of ovariectomized osteoporosis rats,and to elucidate the mechanism of Zuogui Pill in the rat model of ovariectomized osteoporosis,and provide experimental for clinical treatment.Material and method:Eighty Wistar female rats of SPF grade experiment were randomly divided into4 groups according to the weight of the rats.The normal control group was 20,and the normal feeding was not carried out during the modeling period.During the treatment,saline was administered by the stomach.In the other three groups,the bilateral ovaries of the rats were removed for postmenopausal osteoporosis modeling.20 rats in the model group were treated with normal saline during the treatment;20 rats were treated with Bushen Zhuangjing Zhuanggu Chinese Medicine Group(Zuoguiwan group),and Zuogui Pill Solution was used for intragastric administration;alendronate sodium control group(Fushanmei group),20 rats were treated with alendronate sodium solution during the treatment.The treatment period is 60 d.After the treatment,the left femur and tibia of the rats were removed.The bone mineral density of the rats was detected by dual energy X-ray absorptiometry.The rat uterus was extracted,the wet weight index of the uterus was calculated,and the abdominal abdomen of the rats was extracted.Arterial blood,serum was separated,and serum SOD,NO,GSH-Px,and MDA levels were detected.Results:1.After 12 weeks of gavage,the bone mass density of the femur,the first 1/3of the femur and the left lower extremity was significantly lower than that of the normal control group(P<0.05),and the bone density of the tibia was decreased,but the difference was not significant(P> 0.05),suggesting that the bilateral ovaries of the rats can successfully replicate the postmenopausal osteoporosis model;2.After 12 weeks of gavage,compared with the normal group,the body weight of the model group was significantly increased(P<0.05),the change of Fushanmei group was not obvious(P>0.05),and the weight loss of Zuoguiwan group was significantly different(P<0.05).Compared with the model group,the Zuoguiwan group decreased significantly(P<0.05),and the difference was statistically significant;3.After 12 weeks of gavage,compared with the normal group,the wet weight of uterus in Fushanmei group,Zuoguiwan group and model group decreased significantly(P<0.05),and the change in model group was the most obvious,suggesting postmenopausal osteoporosis model rats.The uterus is atrophied.4.After 12 weeks of gavage,the MDA level of the model group was significantly higher than that of the normal control group(P<0.05),the SOD content decreased significantly(P<0.05),and the GSH-Px and NO levels decreased significantly(P<0.05).<0.05),suggesting that the rat’s antioxidant capacity decreased after modeling;5.After 12 weeks of gavage,the left 1/3 and the femoral bone density of the femur and femur were significantly increased in the Zuoguiwan group and the Fushanmei group(P<0.05).The serum SOD content was significantly increased,and the difference was significant(P<0.05),MDA showed a downward trend,no significant difference(P>0.05);serum GSH-Px,NO levels increased significantly(P<0.05),suggesting that Zuoguiwan group and Fushanmei group can improve postmenopausal bone.Antioxidant capacity of rats with osteoporosis.6.After 12 weeks of gavage,the Zuogui Pill group and Fushanmei group were more significant in the left 1/3 of the femur and femur in the Zuogui Pill group(P<0.05).The serum MDA and SOD levels were observed.There was no significant difference(P>0.05),but the detection value of Zuoguiwan group was better than Fushanmei group;the difference of serum GSH-Px and NO levels was not significant(P>0.05),but the value of Zuoguiwan group increased.The magnitude is higher than the Fushanmei group.It is suggested that Zuoguiwan group has better effect on the regulation of antioxidant capacity in postmenopausal osteoporosis rats.Conclusion:1.Zuogui Pill intervention in ovariectomized osteoporosis rats can significantly improve bone density.2.The ovariectomized osteoporosis model has lipid peroxidation damage and decreased antioxidant capacity.3.One of the mechanisms of Zuogui Pill is to prevent and treat osteoporosis by reducing lipid peroxidation damage and improving antioxidant capacity. |
