BackgroundThe incidence of Nonalcoholic fatty liver disease(NAFLD)boosted year by year,and the mechanism of it is not clear now.NAFLD,a kind of metabolic stress-induced liver disease,goes hand in hand with genetic susceptibility and insulin resistance(IR).NAFLD is regarded as the hepatic manifestation of metabolic syndrome(Met S).As a ligands activated transcription factor,Peroxisome proliferator-activated receptorγ-γ(PPAR-γ)has many biological effects,and the Single nucleotide polymorphism(SNP)of PPAR-γis closely associated with Met S and NAFLD.ObjectiveThe aim of this study was to investigate the distribution of PPAR-γ gene rs3856806 polymorphisms and the relationship between non-alcoholic fatty liver disease(NAFLD)in Qingdao han population.MethodsPPAR-γgenotypes of blood samples randomly collecting 189 B-ultrasound-proven NAFLD patients and 183 healthy controls were analyzed by multiple heat ligase detection reaction technology(iMLDR).Distribution of the genotypes of PPAR-γgene rs3856806 polymorphisms was in accordance with the Hardy-Weinberg(H-W)equilibrium in the case and control groups.We compared the differences of the clinical data and the serum lipid profiles determined by biochemical methods,as well as the differences of genotype and allele frequencies between NAFLD and control groups.In this study,the statistical analysis package SPSS 17.0 was used for the statistical analysis.Pearson chi-square test and t test were used for enumeration data and measurement data respectively.The odds ration(OR)and 95% confidence interval(95% CI)was calculatedvia the logistic regression analysis.ResultThe levels of body weight,HC,WC,ALT,AST,GGT,GLU,TG,TC,and LDL were higher in NAFLD group than the controls,and there were significantly different among them(P<0.05).However,the levels of HDL was lower than controls(P<0.01),and the frequencies of PPAR-γ gene rs3856806 allele and genotype were not different between the two groups(P>0.05).The study revealed an increased risk of developing NAFLD for PPAR-γ gene rs3856806 T allele compared with C allele(OR = 1.068,95% CI: 0.748-1.525,P >0.05).Logistic regression analysis indicated that the OR of carriers with the TT + TC genotype of PPAR-γrs3856806 developing NAFLD were1.077,(95% CI : 0.382-1.709,P > 0.05),when compared with those with CC genotype.ConclusionsThe polymorphisms of PPAR-γ gene rs3856806 is not associated with risk of NAFLD in Qingdao han population.
