ObjectiveKusnezoff Monkshood Root was a common poisonous traditional Chinese medicine,but its quality was instability because had no standardization processing technology.To solve the problem of unstable quality of Kusnezoffii and provide a scientific basis for the development of standardized processing technology,we studied inheritance and standardized application of processing methods obout Kusnezoff Monkshood Root.MethodsFocusing on the purpose of these studies,firstly,we roundly refered to the ancient,modern and contemporary literature,and sorted the literature on ancient and modern processing history,chemical composition,pharmacology and toxicology,the study of modern processing technology,quality standards research,clinical research.This could lay a solid foundation for post-experimental study.Secondly,we prepared different processed products,checked the shape-property quality and used HPLC to determine the content of mesaconine,aconitine,hypaconitine,benzoylmesaconine,benzoylaconitine,benzoylhypaconitine in all sample.Comparing the difference of traditional quality and content of alkaloids in products,to explain the scientific connotation of traditional processing experience:removing stem,parting,soaking to no dry heart,cooking until no white.Thirdly,atmospheric pressure boiling,high-pressure boiling and high-pressure steaming cooked different processing time of processed products,checked the shape-property quality and determined the content of six kinds of alkaloids in order to find indicators for process control.Fourthly,in this experiment,we used optimized atmospheric pressure boiling process to prepare 10 batches of pilot scale test sample,checked the shape-property quality,determine the moisture and the content of six kinds of alkaloids.Fifthly,This study determined the content of six kinds of alkaloids in 6 batches Aconitum and 10 batches processed products by Pharmacopeia method and analysis differences between content of alkaloid determined and pharmacopoeia quality standards.Lastly,Reference to the pharmacopoeia processing methods,we prepared different concentrations of vinegar processed products,checked the shape-property quality and determined the content of six kinds of alkaloids.ResultFirstly,cleaning is to remove the non-medicinal parts whose content of six alkaloids is low;after tranching by density,processing Aconitum can save hours of work,make quality uniform and reduce the loss of the active ingrediengt;when Aconitum was soaked moderately,it was in favour of cuting into slices and the quality of decoction pieces was good;when Aconitum was cooked until no white,the content of three kinds of diester alkaloids in Aconitum conformed to the pharmacopoeia requirements,the change of content of three kinds of monoester alkaloids in Aconitum was moderate and the quality of decoction pieces was good.Secondly,when Aconitum was cooked 4~6h in atmospheric pressure,its traditional quality and content of diester alkaloids meet the requirements and its content of ester alkaloids changed smooth.So,We initially identified that atmospheric pressure cooking time is 4~6h.In addition,high-pressure cooking method needed less working hours than atmospheric pressure cooking.Thirdly,there was no significant differences in traditional quality,moisture(P>0.05)and six kinds of alkaloids(P>0.05)of 10 batches Aconitum.The range of total content of monoester alkaloids in ten batches Aconitum was 0.188%~0.247%and was much higher than the Pharmacopoeia standards that the content of monoester alkaloids was 0.020%~0.070%.Fourthly,The range of total content of diester alkaloids in 6 batches Aconitum was 0.412%~0.731%and the content of diester alkaloids in four batches Aconitum were higher than the Pharmacopoeia standards that the content of diester alkaloids was 0.10%~0.50%.The range of total content of monoester alkaloids in ten batches Aconitum was 0.162%~0.446%and the content of monoester alkaloids in all were much higher than the Pharmacopoeia standards that the content of monoester alkaloids was 0.020%~0.070%.Lastly,vinegar may reduce outflow of alkaloids in the soaking process,inhibit the hydrolysis of diester alkaloids in the cooking process.The concentration of vinegar was higher and the inhibitory was stronger.Effects of vinegar on other ingredients in Aconitum and efficacy will be studied in the future.ConclusionIt is meaningful of traditional processing experience when processing Aconitum at atmospheric pressure.When traditional quality meet requirements,diester alkaloids was lower and monoester alkaloids changed smooth,the processing time can be used as quantitative indicators to control quality of Aconitum.The quality of Aconitum which processed by preferred processing technology was basic stability,that provided a scientific basis for the establishment of SOP.We believed that the pharmacopoeia quality standard of Aconitum processed products should be improved.It was meaningful of controlling the quality of Radix Aconiti Kusnezoffi preparata.
