Objective Based on analysis of flow balance(flux balance analysis, FBA) method of metabolic network modeling and analysis platform, the application of the project is to establish coronary heart disease, myocardial cell energy metabolism disease of blood stasis syndrome network model, to carry out related experimental research,to verify the practicability and validity of network model disease.Methods Step 1: copy of the animal models of chronic myocardial ischemia of rats; Step 2: To Simulation and application the coronary heart disease of blood stasis syndrome myocardial cell energy metabolism.Disease syndrome network model. After the building modes of rats,Compared with the healthy rats,blood lipid, and blood hemorheology.Each index showed increased significantly,The difference was statistically significant,The ECG ST segment significantly higher. That has been successfully copied chronic myocardial ischemia model in rats.2.Three days,seven days and ten days isopropyl adrenaline injection group compared with healthy controls,the myocardial cell energy metabolism material: fatty acids,O2 and ATP value compare the difference was statistically significant.3. Compared with the healthy group, ischemia group of blood stasis syndrome rats serum metabolomics differences metabolites have glycolic acid, pyruvic acid, valine, isoleucine, Nmethyl DL- pyruvic acid, mannose glyceric acid.4. After the treatment of Yangxin tongmai tablets, the bottom of the myocardial cell energy metabolism: fatty acids, O2 and ATP value comparison, compared with healthy controls, the three indicators blank model group was obviously lower. The difference was statistically significant. Compared with the model group, yangxin tongmai tablets group, the trimetazidine group increased significantly, the difference was statistically significant; Compared with yangxin tongmai tablets group, trimetazidine group value is low. The difference was statistically significant.Conclusions: 1.The application of high-fat feeding,vitamin D3,isopropyl epinephrine injections could successfully made chronic myocardial ischemia model in rats. 2. In the condition of myocardial ischemia, myocardial cell energy metabolism pathways damage, decrease ATP production, energy supply. 3. Flow balance analysis indicates that the healthy controls, mild ischemia group, moderate ischemia group, severe ischemic myocardial cell energy metabolism in group of four groups of ATP production will decrease trend, it has to do with the key enzyme of the Krebs cycle in four groups(Cs, concentration of Icdh, Od) change is consistent, build the energy metabolism of blood stasis syndrome of coronary heart disease can network model accurately predict coronary heart disease to some extent under different degree of ischemia state energy substrate metabolism, the change of key enzyme and ATP production. 4. The experiment based on flow balance analysis software(flux balance analysis, FBA),combined with the animal experiments,the preliminary simulate normal rat myocardial cell energy metabolism network model,and confirmed the yangxin tongmai tablets can alleviate myocardial ischemia effectively,improve the function of the heart blood supply,curative effect is more significant in the trimetazidine group.
