Macleyea cordata(Willd)R.Br., a plant belonging to genus of Macleaya, Papavcreraceae family, is a perennial tall erect herb, mainly distributed in the south provinces of Yangtze river in China. This plant has effects of antibacterial, antiinflammatoary, insecticidal, and so on. Previous studies reported that the mainly bioactive components were alkaloids.This paper, we mainly studied the alkaloids components in fresh roots of M. Cordata. After extraction, concentration and roughly separation, finally got 7 fractions, F1-F6. Every fraction was analysed by LC-MS, the molecular weight of the compounds were obtained, and the main alkaloids were existed in F3. More over, the structures of some compounds were preliminarily determined by MS/MS, this has been extremely helpful in finding unknown alkaloids quickly in M. Cordata. As a result, we got 20 compounds(19 alkaloids, 1 sterol) by kinds of chromatographic ways, including silica gel column chromatography, recrystallization, preparation thin layer plate and HPLC, 1 new compound, 2 new natural compounds, 4 compounds were isolated from M. Cordata for the first time.The structure of the 19 compounds were identified by spectroscopic metho ds, MS, 1H-NMR, 13C-NMR, 2D-NMR, DEPT. The result as follow: dihydrosan gunarine(1), dihydrochelerythrine(2), dihydrochelirubine(3), 6-cyanodihydrosangu narine(4), 6-acetonyldihydrochelerythrine(5),sterol(6),(R)-6-((R)-1-hydroxyethyl)di hydrochelerythrine(7),(S)-6-((R)-1-hydroxyethyl)dihydrosanguinarine(8),(S)-6-((R)-1-hydroxyethyl)dihydrochelerythrine(9),(2-hydroxyethyl)dihydrochelerythrine(10),6-c yanodihydrochelilutine(11),6-Cyanodihydrochelerythrine(12), 8-demethyl-dihydroch elerythrine(13), cheilanthifoline(14), 6-methoxy-norchelerythrine(15), canadine(16),sangunarine(17), chelerythrine(18), protopine(19), allocryptopine(20). Compound10 was new compound, compound 11 and 12 were new natural products, com pound 13, 14, 15 and 16 were isolated from M. Cordata for the first time.
