| Objective To explore the acid-sensing ion channels(ASIC1a)expression in mice airway tissue,and investigate the level of intracellular calcium ion concentration changes and apoptosis induced by acidification in EBTr cells.Methods The expression of ASIC1a mRNA and protein in mice airway tissue was evaluated by real-time polymerase chain reaction(PCR),Western blotting and immunohistochemistry.The experiment is divided into three groups:normal group,acidification group(pH6.0),specific blocker(PcTx1)group.Observing the intracellular calcium([Ca2+]i)changes of ASIC1a through flo-3,and the apoptosis of EBTr cells through DAPI staining and Annexin V-FITC/PI flow cytometry in each group.Results1.Detected the expression of ASIC1a mRNA and protein in mice airway tissue.2.Extracellular pH6.0 increased[Ca2+]i in the existence of extracellular Ca2+,pharmacological intervention with the ASIC1a-specific blocker PcTx1 venom significantly reduced this increase in[Ca2+]i.3.Apoptosis increasedinpH6.0groupandPcTx1group had no obvious responses to acidification.Conclusion The ASIC1a expresses in the respiratory tract of mice,extracellular acidification increased the[Ca2+]i level and the apoptosis in EBTr cells. |
