| Objective:To investigate the effect of acute ethanol intoxication(AEI)on the expression of GFAP and AQP4 in serum and brain tissue after traumatic brain injury(TBI)in rats.The underlying mechanism was also investigated.Method:45 rats according to body weight were randomly assigned to 3 groups,as experimental group 1,2 and control group,each group has 15.Experiments of 1,2 group rats were given ethanol(ethanol diluted with distilled water)concentration was 30%,60%by 1 ml/100g liquid irrigation stomach,the control group was given the same dose of distilled water.1 h after gavage of rats in each group by improved Feeney s free falling epidural hit method for traumatic brain injury model.(1)After TBI 1 h,6 h,24 h,respectively from each experimental group and control group were randomly selected from five rats blood,subsequent high-speed centrifugation,the levels of GFAP and AQP4 in serum was determinated.(2)The rats were killed after blood was drawn and the brain was quickly removed,and the levels of GFAP and AQP4 in brain tissue was determined.Results:(1)Compared with the control group,the expression of AQP4 and GFAP in serum and brain tissues of 1,2 group were significantly increased(P<0.05).(2)The expressions of AQP4 and GFAP in serum and brain tissues of the 2 groups were significantly higher than those in the 1 groups(P<0.05).(3)The expression of AQP4 and GFAP in the serum and brain tissue of the three groups was more obvious with the prolonging of the time of injury(in 24h).Conclusions:Acute ethanol intoxication can increase the level of TBI in rats,and the higher the concentration of ethanol in blood,the longer the injury,the more severe the TBI. |
