Objective:To investigate the effect of Mannitol on brain water content and AQP4 expression of traumatic site and hippocampus in severe traumatic brain injury(TBI) and the relationship between the brain edema and AQP4 expression.Methods:Twenty-four mice were randomly divided into three groups:TBI group,sham operation group and TBI+Mannitol(experimental group). The brains of the animals were taken out 8h after TBI. The expressions of AQP4 were detected by using immunohistochemistry. The wet-dry weighing method was used to detect the brain water content.Results:Brain water content of Experimental group 8h after TBI was lower than that of TBI group, but higher than that of the sham operation group. The expression of AQP4 of the experimental group was lower than that of TBI group, but higher than that of sham operation group.Conclusion:AQP4 may have close relationship in the formation and development of brain edema, and play an important role in the exploration of new dehydration drug in treatment of brain edema and the relationship between hippocampus and traumatic brain injury.
