Background:Nowadays, about 35%to 50%of the patients with traumatic brain injury(TBI) accompany ethanol intoxication. And acute ethanol intoxication(AEI) probably leads to a poorer outcome in these patients with head trauma. This study is planned to analyze the influence of AEI on the apoptosis after severe TBI as well as the related gene expression.Methods:48 Sprague-Dawley rats were assigned randomly to TBI group(n=24) and TBI+ AEI group(n=24).All SD rats were subjected to severe TBI following Feeney's method, however,in TBI+ AEI group, before head trauma, the animals were subjected AEI first. In each group animals are sacrificed 6h,24h,72h,168h after TBI. Apoptosis and expression of Bcl-2 were determined by TUNEL and immunohistology relatively. Results:Microscope examination showed that intensive distribution of apoptosis cells could be seen in the brain tissue circulated the injured cortex.Apoptosis cells were determined just 6h post head trauma, and reached peak at 72h.The rate of neurocyte apoptosis in TBI+ AEI group after TBI was higher than TBI group, and the expression of Bcl-2 was higher as well. Conclusion:Our findings suggest that AEI would increase neurocyte apoptosis after severe TBI,and would change the expression of related apoptosis gene...
