Effect Of Shuang Hong Qin Yu Tang Decoction In The Concent Of IL-1β And NF-KB On Rat With Ulcerative Colitis

Objective: ulcerative colitis is a kind of recurrent intractable chronic nonspecific inflammatory bowel disease.This experiment is by observing the result of Hua Zhuo Jiedushuanghongqinyutang to the ulcerative colitis rats′general condition,the disease activity index(DAI),colonic mucosal tissue pathology change,the level of interleukin 1β(IL-1β)in the serum,the content of NF-ΚB in the colonic mucosal tissue,to explore the mechanism of Hua Zhuo Jiedushuanghongqinyutang to the ulcerative colitis and to provide experimental basis for clinical treatment.Methods: 60 healthy male Wistar rats weighting at 180g-250 g were randomly divided into 6 groups,each group of 10,respectively Normal Control Group(NK),Model Group(M),Mesalamine Group(MSG),Shuang Hong Qin Yu Tang High Dose Group(HG),Middle Dose Group(MG),and Low Dose Group(LG).After 1 week of adaptive feed,NK group was breeding as usual,the rest of the groups of rats with 2 minus 2-trinitrobenzene sulfonic acid(TNBS)/ethanol joint big rats UC model building method.Observing four days later,on the basis of randomly,1 rat from each group(except NK),observing the rats are in low spirits,bloody purulent stool,which reminded building success.Then NK group and M group continue to normal raise,MSG group was given Mesalamine lavage,HG,MG and LG group were given high,medium and low three concentrations of Shuang Hong Qin Yu Tang lavage.During the period of treatment,the rats body weight,hair color,mental state,shit character were observed,then we give them DAI score.The treatment totally continued 1 weeks.After the treatment,only water giving for24 hours,then killing the rats,keeping the blood to determined the levels of IL-1β,and cutting the most obvious in colon mucosal tissue to detect the content of NF-KB.Results:1 general observation and DIANK group rats were in good condition.After building,the rest of the groups of rats′ depression,love nuzzles,food intake and activity were significantly decreased,cluster,unresponsive,back arched,hair loose dumb yellow without luster,weight decreased obviously,waste dilute or mucous purulent blood.After treatment,the treatment group rats also had an improved in general,the HG group improved the most obvious,MSG,MG group is no significant difference,were slightly high in LG group,slightly less in HG group.DAI score results showed that HG(0.410±0.283),MG group(0.889±0.367),LG group(1.323±0.251)and MSG group(0.908±0.290)were lower than the M group(2.479±0.244),statistically significant difference(P<0.05);the scores of the HG was lower than that of the MG,MSG and LG group,statistically significant difference(P<0.05);MSG group compared with the MG group,there was no statistically significant difference(P>0.05).The scores of the MSG group and MG group were lower than the LG group,statistically significant difference(P<0.05).2 colonic mucosa tissue pathological characteristicWhen we observed the colonic mucosal tissue of each group by Naked eye,we found it is of hyperemia edema in each group(except NK),and M group is the most severe.From severe to mild in turn is LG group,MG group,HG group.There was no statistically significant difference between MSG group and MG group.The colon mucosa tissues of NK group rats were normal.The colon mucosal ulcer of M group rats were most,glandular structure disorder,annular plica disappear,hyperemia,edema obvious.HG group has superficial ulcer,and found amounts of glands freshmen;MG group,MSG mucosa owe smooth,a small ulcer wound;LG group mucosal ulcer slightly more visible,glandular structure serious damaged,less new glands,mucosal hyperemia,edema,plasma cells,lymphocytes,such as cell infiltration.3 IL-1β content and determination of the content of NF-ΚBThe contents of L-1β in NK(0.109±0.005),HG(0.139±0.006),MG group(0.171±0.007),LG group(0.172±0.009)and MSG group(0.152±0.007)were lower than that of M group(0.226±0.028),statistically significant difference(P<0.05);HG group and LG,MG group and MSG group,the difference is statistically significant(P<0.05);NK group and MSG group,HG group,MG group,LG group,the difference is statistically significant(P<0.05);MG and LG group,there was no statistically significant difference(P>0.05).NF-KB: NK(0.030±0.011),HG group(0.052±0.010),MG(0.117±0.007),LG group(0.140±0.007),and MSG(0.080±0.011)content is lower than that of M group(0.179±0.024),the difference had statistical significance(P<0.05).MG,LG group,HG group and MSG content,the difference had statistical significance(P<0.05).HG,MG,LG and MSG group content is higher than that of NK group,the difference had statistical significance(P<0.05).HG group,MG group and LG group,the difference was statistically significant(P<0.05).Conclusion:1 Shuang Hong Qin Yu Tang can improve the general situation of ulcerative colitis rats,repair the injury colon mucosa,at the same time reduce the DAI score,and present a certain amount-effect relationship,of all these groups,the effect of HG is the most.2 Shuang Hong Qin Yu Tang can improve the general situation of ulcerative colitis rats,repair the injury colon mucosa,its mechanism might be down regulating the content of inflammatory cytokines IL-1β,inhibiting inflammation,down regulating NF-KB content to promote restoration of the mucosa.
