Objectives The oxysterol related protein-9(Oxysterol Binding Protein-related Protein-9,ORP-9)as a candidate gene of cerebral infarction and its detection in Hebei Han population frequency distribution,analysis of the relationship between ORP-9 gene polymorphism and cerebral infarction,and analyze the key of atherosclerosis and risk factors of cerebral infarction-AS(Atheroselerosis),the relationship between blood pressure and the Blood lipids and blood sugar.Methods In the study we chose 174 patients from Affiliated Hospital of North China University of Science and Technology with cerebral infarction as CI group,and the age and sex-matched individuals were chosen 117 as the control group,these individuals were from the same hospital without cerebral infarction or North China Polytechnic University Affiliated Hospital Medical Center with healthy.Blood samples of 5ml were collected in the early morning of the study,and the patients were followed up for their history and related tests,we took case-control study with the control group.We extracted fasting venous blood and DNA from whole blood.Then we used PCR testing,the product of PCR were used Restriction fragment length polymorphism(RFLP)to detected single nucleotide polymorphism of ORP-9 two sites rs856600 and rs768529,and we tested the the genotypic allelic distributions.T test,chi square test and logistic regression model were used to deal with the statistical data,and the statistical data was analyzed by using SPSS l 7 statistical software.Results 1 In the comparison of basic clinical data of cerebral infarction group and control group,in many aspects,such as age,sex,smoking and drinking history,there is no statistical significance between the two groups(P>0.05);when compared with the control group,cerebral infarction group on BMI,diabetes meilitus,hypertension,systolic blood pressure,diastolic blood pressure,fasting blood glucose,Triglyeeride,Total cholesterol,and Low gensity lipoprotein(P<0.05).2 Rs856600 and rs768529 genotype and allele frequencies were not statistically significant in the cerebral infarction in comparison with the control group(P>0.05);the results of logistic regression analysis does no correlation between the frequencies of all genotypes and alleles and the incidence of cerebral infarction(P>0.05).3 There was no significant difference in genotype and allele frequency distribution between the two sides of ORP-9 in patients with cerebral infarction(HBP+CI)and those with no history of hypertension(HBP+CI)(P>0.05).There was no significant difference in the genotype and allele frequencies of the two ORP-9 loci in patients with cerebral infarction(DM+CI)and those without diabetes mellitus(DM+CI)(P>0.05).4 Blood pressure,fasting blood glucose,and TG,TC,HDL,and LDL levels were not statistically significant in the comparison between rs856600's C and T(CT+TT)and rs768529's A(AA+AT)and T populations(P>0.05).5 The genotype and allele frequencies of rs856600 and rs768529 were not statistically significant in WAS combined with cerebral infarction and WAS control group(P>0.05).The genotype and allele frequencies of rs856600 and rs768529 were not statistically significant in AS combined with cerebral infarction and AS control group(P>0.05).The genotype and allele frequency of rs856600 was not statistically significant in VP combined with cerebral infarction and SP combined with cerebral infarction(P> 0.05).The genotype and allele frequency of rs768529 was correlated with VP combined with cerebral infarction and SP combined with cerebral infarction(P <0.05).Conclusions The genetic polymorphism of two ORP-9 sides may not be associated with cerebral infarction and the risk factors of cerebral infarction such as hypertension,hyperglycemia and TG,TC,HDL,LDL and other blood lipid factors in Hebei Han population.The genotype and allele frequencies of rs856600 were not associated with AS,VP and SP(P>0.05),and the rs768529 allele of T may be a risk factor for vulnerable atherosclerotic plaques in Hebei Han population.
