| Objectives: Through the establishment of ulcerative colitis rats model,to observe the of Herba Houttuyniae with Dragon’s blood on serum IL-10、TNF-α、PAF on UC Model Rats,to verify Herba Houttuyniae with Dragon’s blood for the treatment of UC and to investigate weather the Herba Houttuyniae with Dragon’s blood is synergy and its mechanism of action for the treatment of UC.Methods: Adaptability to 48 male and 48 female SD rats fed a week later,randomly selected 12 for blank group,the other to build module.before building,All rats fasting and drink 24 h,free drinking water.Trinitrobenzene sulfonic acid(TNBS)/ ethanol solution made after the success of the model,model of rats were randomly divided into 7 groups,namely modeling group,salazosulfapyridine(SASP)group,Herba Houttuyniae with Dragon’s Blood high dose group,Herba Houttuyniae with Dragon’s Blood middle dose group,Herba Houttuyniae with Dragon’s Blood low dose group,Dragon’s Blood group,Herba Houttuyniae group.Each group were given corresponding drug lavage,3ml/second,1 time/day.During the dosing,observed every groups of general condition,body weight change and fecal occult blood test.After last time dosing,every group fast but drink 24 h.At abdominal cavity anesthesia heart blood,all rats were executed by drawing-out the blood in their heart,and detected of serum IL-10、TNF-α、PAF level.After the blood using the method of cervical dislocation to kill the rats and taken colon tissue to observed the change of the colonic mucosa and make pathological examination.Results: Herba Houttuyniae with Dragon’s Blood high dose group than Herba Houttuyniae with Dragon’s Blood middle dose group、SASP group、Dragon’s Blood group、Herba Houttuyniae group、Herba Houttuyniae with Dragon’s Blood low dose group、modeling group,the general state of rapid recovery,purulent blood disappears earlier,middle dose group and SASP group have similar curtive effect.Combination groups and SASP group、single drug groups showed mucosal hyperemia edema disappeared,erosion and ulcer healing,partly on the ulcer significantly reparative changes.UC modeling group rats serum IL-10 levels significantly decreased in the blank group,the TNF-α、PAF increased obviously(P<0.01);After gavaging,IL-10 of the Herba Houttuyniae with Dragon’s Blood high dose group was obvious higher,the TNF-α、PAF is decreased obviously,comparing with modeling group、SASP group、the low dose group、the middle group、Dragon’s Blood group and Herba Houttuyniae group,they have significant difference(P<0.01);the Herba Houttuyniae with Dragon’s Blood middle and low dose group、Dragon’s Blood group、Herba Houttuyniae group、SASP group IL-10 levels has also in creased,the TNF-α、PAF fell too,there are significant difference compared with model group(P<0.01).SASP group compared with Herba Houttuyniae with Dragon’s Blood middle dose group、Dragon’s Blood group and Herba Houttuyniae group,there was no significant difference(P>0.05),but compared with Herba Houttuyniae with Dragon’s Blood low dose group,the difference is ststistically significant(P<0.05);Herba Houttuyniae with Dragon’s Blood low dose group compared with middle and high dose group,the difference is statistically significant(P<0.05),but with Dragon’s Blood group and Herba Houttuyniae group,there was no statistically significant difference(P>0.05).Conclusions: 1.The Herba Houttuyniae and Dragon’ Blood have treatment effect to rats UC model,they have synergy,and they are curative effect is better than that of SASP.2.The Herba Houttuyniae with Dragon’s Blood possible mechanism for the treatment of UC:(1)Increase anti-inflammatory factor IL-10,thus inhibiting proinflammatory factor,balancing the intestinal cells,maintaining normal intestinal mucosal immune regulation.(2)Cut the platelet activation PAF,improving the state of platelet activation and promoting tissue repair.Cut proinflammatory factor TNF-α,reduce colon tissue injury of acute inflammation,granuloma formation in block ulcerated lesions and fibrous tissue hyperplasia,thus promoting ulcer healing. |
