Objective1 Studying the distribution regularity of TCM syndromes of chronic obstructive pulmonary disease in the period of acute aggravation2 Studying the influence of SQHTF on MMP-9?TIMP-1?p-MLC and p-MYPT-1 of lung tissue in COPD rats.Methods1 Clinical research:making a TCM syndrome questionnaire of AECOPD by searching relevant literatures to collect the general information and four diagnosis of TCM of 120patient's,the distribution of TCM syndrome of AECOPD was got by data analysis.2 Animal experiment:Rats were divided into normal group and model group based on the randomized table.The rats in the model group were treated with the method of smoking and instillation of lipopolysaccharide into the airway to replicate the COPD model,the rats were randomly divided into 5 groups after the COPD model established successfully:model group,JSB group,three levels of SQHTF groups(high,middle and low).After 28 days of treatment,lung function and tissue samples were collected.Immunohistochemical method was used to detect the expression of MMP-9 and TIMP in lung tissues.Western blot method was used to detect the expression of p-MLC and p-MYPT-1 in lung tissues.Results1 Clinical research:The highest frequency of the five TCM syndromes were Syndrome of Qi deficiency of lung and spleen combined with phlegm heat and blood stasis(20.83%),Syndrome of Qi deficiency of lung and spleen combined with phlegm and blood stasis(15.83%),Syndrome of both Qi and Yin deficiency of lung and kidney combined with phlegm heat and blood stasis(13.33%),Syndrome of Qi deficiency of lung and spleen combined with phlegm(11.67%),Syndrome of Qi deficiency of lung and kidney combined with phlegm heat(8.33%). 2 Animal experiment2.1 Change of pulmonary function Compared with the normal group,the pulmonary function of the other groups was significantly lower,which was statistically significant(P<0.01 or P<0.05).Compared with the model group,the lung function of each treatment group was significantly improved,which was statistically significant(P<0.01 or P<0.05).Compared with each other in the treatment group,the pulmonary function of the high level of SQHTF group was improved obviously,which was statistically significant(P<0.05).2.2 Pathological changes The structure of lung tissue of rats in the normal group was normal,the structure of rats in model group was in accordance with the pathological changes of COPD,which manifested as bronchial stenosis,a large number of inflammatory cell infiltration in the trachea wall,thinning of the alveolar wall,enlargement of the alveolar space,rupture of elastic fibers and fusion of alveoli.The damage of pathological changes of lung tissue was reduced in the both SQHTF groups and JSB group,which reduced most obviously was the high level of SQHTF,followed by the middle level of SQHTF.2.3 The expression of MMP-9 and TIMP-1Compared with the normal group,the expression of MMP-9 and TIMP-1 in the lung tissues of the other groups were significantly higher than those in the normal group,which was statistically significant(P<0.05),and the expression of MMP-9 and TIMP-1 in the model group was the highest.Compared with the model group,the expression of MMP-9 and TIMP-1 in the lung tissues of the rats in each treatment group were decreased,which was statistically significant(P<0.05).Compared with each other in treatment group,the expression of MMP-9 and TIMP-1 in lung tissues of rats of the high level of SQHTF group decreased significantly,which was statistically significant(P<0.05).2.4 The expression of p-MYPT-1 and p-MLC Compared with the normal group,the expression of p-MYPT-1 and p-MLC in the lung tissues of the other groups were significantly higher than those in the normal group,which was statistically significant(P<0.05),and the expression of p-MYPT-1 and p-MLC in the model group was the highest.Compared with the model group,the expression of p-MYPT-1 and p-MLC in the lung tissues of the rats in each treatment group were decreased,which was statistically significant(P<0.05).Compared with each other in treatment group,the expression of p-MYPT-1 and p-MLC in lung tissues of rats of the high level of SQHTF group decreased significantly,which was statistically significant(P<0.05).Conclusion1 Chronic obstructive pulmonary disease in the period of acute aggravation often performed as syndrome of intermingled deficiency and excess.2 SQHTF not only can improve the lung function of COPD rats,but also can relax the bronchus by reducing the expression of p-MYPT-1 and p-MLC,at the same time,reducing lung injury,airway inflammation and airway remodeling by reducing the expression of MMP-9 and TIMP-1.
