Objective: To establish radioactive esophagitis rabbit models, observe the intervention effect ofrabbits with radiation-induced esophagitis which treated by modified zhuye shigao decoction(MZSD), shashen maidong decoction(SSMD) and xuefu zhuyu decoction(XFZY).explore the characteristics of different prescriptions and find them possible mechanisms.Methods: 1.Fifty rabbits,were randomly divided into control group, 16Gygroup, 24Gy group, 32Gy group and 40Gy group.Each group except control group were irradiated by 6MV linear at a dose of 16Gy, 24Gy, 32Gy and 40Gy. The entire esophageal tissues of rabbits were taken for pathology analysis in 7days and 14days afterradiation to make sure the optimal dose for experimental model. The food intake and weight changesof rabbits were observed at the same time.2. Fifty rabbits, clean grade, were randomly divided into control group, model group, SSMD group, XFZY group and MZSD group.All rabbits except those in control group were irradiated by 6MV linear at a dose of 32Gy.The drugs which were given started from one week before the irradiation to one week after the irradiation. After that total esophagus of rabbits in all groups were harvest for detecting. HE staining was used to observe the pathological changes. The contentof nitric oxide(NO), hydroxyl radical(·OH),catalase(CAT) and glutathione peroxidase(GSH-PX) were measured by colorimetry.The level of Bcl-2 mRNA and Bax mRNA expression in the esophagus tissue were measured by RT-PCR and the level of Bcl-2 protein and Bax protein expression by Western blotting.Result:1.Thepathological damage of radiation esophagitis grew as the increase of dose. 32Gy radiation dose group got the highest damage score with no rabbits died(P<0.05). There was no significantdifference in pathological lesion between 7 and 14 days (P>0.05)after rabbits radiated by 32Gy X-rays. 7-9days after irradiation, the daily diet and weight of rats dropped to the lowest level.2. The pathological score of MZSD group was lower significantly than the SSMD group and XFZY group(P<0.05). The content of NO, CAT and GSH-PX of all three drug given groups decreased significantly compared with model group, while the inhibition capacity of·OH increased(P<0.05).The MZSD group performed better than the SSMD group and XFZY group in reducing the content of NO and increasing the inhibition capacity of ·OH(P<0.05). The expressions of Bcl-2 mRNA and protein of MZSD group increased while the expressions of Bax mRNA and protein decreased(P<0.05), compared with the model group. The value of Bcl-2/Bax of MZSD group increasedsignificantly (P<0.05), compared with the other two groups.Conclusion:Radioactive esophagitis rabbit models can be established after 7days using 6MV X-rays in 32Gy irradiation.Compound zhuye shigao granule(MZSD) can significantly reduce the pathological grades and lower oxygen radical level , increase the level of Bcl-2 mRNA and protein expression, decrease those of Bax.It had good prevention effect for acute radiation-induced esophagitis. The effect was better than zhuye shigao decoction and xuefu zhuyu decoction. |
