ObjectiveTO explore the relationship between the variable frequency electroacupuncture?EA?and the efficacy of Vascular dementia?VaD?by contrasting the relative expression of AVP and A?1-40mRNA in Hippocampus CA1 area and the results of Morris Water Maze experiment.Then explore the mechanism of electroacupuncture in the treatment of frequency conversion vascular dementia and choose the better stimulation.Methods120 Adult male SD rats were randomly divided into 6 groups——normal group,sham operation group,model group,piracetam group,2/100Hz EA group and2/15Hz EA group.Normal group without surgical operation as blank control group.Sham operation group for operation,but just need to find the big rat carotid artery and vertebral artery on either side of the hole.The rest of the rats using four-vessel occlusion 4-VO Blocking to make vascular dementia model:first,find the hole on both sides of the vertebral artery of rats,with electric coagulation block;then find the either side of the carotid artery carring on the reversibility of the clip.After the surgery,using Morris water maze screening VaD model and dividing the successful model rats into four groups:VaD model group?N=12??piracetam group?N=12??2/100Hz EA?N=12??2/15Hz EA?N=12?,VaD model group be left untreated,2/100Hz EA and 2/15Hz EA take the electric acupuncture on "dazhui","baihui",electric acupuncture therapy for:treatment once a day,continuous treatment for five days,,then rest two days.Both the two electric acupuncture groups choose 3 mA scheme and dilatational wave,above two courses,while the difference is 2/100Hz EA with 2/100Hz,2/15HzEA with 2/15Hz.Piracetam group with its Suspension lavage treatment.After the treatment,use the Morris water maze to test the learning and memory ability among the treatment groups;at last remove the hippocampus of rats on the ice bags,Using the real-time fluorescent quantitative PCR detection to test the difference of the relative expression level of AVP and A?1-40 mRNA in Hippocampus CA1 area.Results1.Morris water maze resultsCompared with others groups,the mean escape latencies and the first time of crossing platform in model group were significantly longer?P<0.05,P<0.05?and the times of crossing the original platform quadrant within120seconds was markedly less?P<0.05?.Compared with EA groups,Piracetam group has no statistical difference with P>0.05.Comparison between 2/100Hz EA group and 2/15Hz indicated that the escape latency and the first time to crossing platform in the later group was markedly shorter than that of the former group?P<0.05?,which also has more times to crossing the original platform quadrant within120 seconds?P<0.05?.2.Fluorescence PCR test resultsCompared with others groups,the gene expression level of AVP which from Hippocampus CA1 area in model group was significantly lower than the others?P<0.05?.Compared with EA groups,the AVP expression of Piracetam group has no statistical difference?P>0.05?.Comparison between two EA groups,2/15Hz EA group had a higher expression level than 2/100Hz EA group?P<0.05?.Compared with others groups,the gene expression level of A?1-40mRNA which from Hippocampus CA1 area in model group was significantly higher than the others?P<0.05?.Compared with EA groups,the A?1-40mRNA expression ofPiracetam group has no statistical difference?P>0.05?.Comparison between two EA groups,2/15Hz EA group had a lower expression level than 2/100Hz EA group?P<0.05?.Conclusion1.Both electroacupuncture and Piracetam can improve the ability of learning and memory in VaD rats,the mechanism which perhaps are the little lower expression level of A?1-40mRNA and the higher expression level in Hippocampus CA1 area.2.Effects from different variable frequency electroacupuncture on VaD rats had significant differences,which were inclined to 2/15Hz EA beng better than 2/100Hz EA.
