Objective: To observe the effect of roubin manipulation’s on knee osteoarthritis in rabbits influence the model of bone tissue morphology,serum matrix metalloproteinase MMP-3,MMP-9,MMP-13,MMP-14 and its inhibitor TIMP-1 content change,further deepen our understanding of KOA pathogenesis,to explore the mechanism of roubin manipulation’s in the treatment of KOA,provide the basic theory for the prevention and treatment of KOA.Methods: 52 New Zealand rabbits were randomly divided into manipulation’s group,control group,sham operation’s group and the normal group(n=13).manipulation’s group and control group were established into high intraosseous pressure experiment model of right leg caused by ligating the inferior gluteal vein,femoral vein and great saphenous vein,sham operation’s group surgical exposed but not the corresponding vascular ligation;normal group without any treatment.One week later,manipulation’s group was treated with roubin manipulation’s for 5 consecutive weeks,A total of 17 treatments.the remaining three groups without any treatment.At the end of the 1 weeks and 12 weeks,The content was measured of serum MMP-3,MMP-9,MMP-13,MMP-14 and TIMP-1 in rabbit of ear vein blood by ELISA method.At the end of the 12 week to take the right knee medial tibial plateau cartilage tissue were observed by macroscopic observation and light-microscopic of articular cartilage and accorded with the modified Mankin grade standard for evaluation of pathological score.Results: 1.At the end of the 12,after observed by macroscopic observation and light-microscopic of articular cartilage.We found:manipulation’s group:cartilage was blue,visible surface cracks,four layer structure is clear,some animal cartilage layer thickening,surface layer and middle layer cells increased;the control group:cartilage yellow and dull,rough surface,visible erosion and see the surrounding cartilage defect,tufted hyperplasia,thinning of the cartilage layer significantly,cartilage cells decreased significantly;sham operation’s group:fracture cartilage surface was irregular,some animal cartilage fracture depth of transitional layer,the four layer of cartilage structure is clear,the number of cartilage cells is normal;normal group: cartilage surface smooth cartilage,four layer structure is clear,the number of cartilage cell is normal.Compare with the cartilage tissue pathology ManKin score of manipulation’s group,control group,sham operation’s group and normal group increased significantly compared to that,with statistical difference(P<0.05).The control group animal cartilage tissue pathological observation and ManKin score of moderate degeneration,manipulation group and sham module with mild degeneration,the normal group had no obvious degeneration.2.At the end of the first week,manipulation’s group compared with control group,there was no significant difference in the levels of serum MMP-3,MMP-9,MMP-13,MMP-14 and TIMP-1(P>0.05).The MMP-3,MMP-9,MMP-13,MMP-14 and TIMP-1 levels were significantly higher in the control group compared with sham operation’s group and the normal group,the difference was statistically significant(P<0.05).The control group was compared with sham operation’s group and the normal group,the indexes were significantly increased,the difference was statistically significant(P<0.05).Compared with the normal group,the contents ofMMP-3,MMP-9,MMP-13,MMP-14 and TIMP-1 were no significantly different between sham operation’s group and the normal group(P>0.05).At the end of the 12 th week,the,the contents of serum MMP-3,MMP-9,MMP-13,MMP-14,and TIMP-1 in the control group were significantly higher than those in manipulation’s group,the difference was statistically significant(P<0.05).The MMP-3,MMP-9,MMP-13,MMP-14 and TIMP-1 contents in the sham operation’s group and the normal group at twelfth weeks were no significant difference than those at the end of the first week,which was no statistically significant(P>0.05).At the end of the twelfth week,control group compared with manipulation’s group MMP-3,MMP-9 and MMP-13,MMP-14 content increased,the difference was statistically significant(P<0.05),but the TIMP-1 content of manipulation’s group is increase compared with the control group,that was a significant difference(P=0.03).The control group was significantly higher than the sham operation’s group and normal group,the difference was statistically significant(P<0.05).Compared with the sham operation’s group and the normal group,the contents of each index in the manipulation group were significantly increased,which could be considered to be statistically significant(P<0.05).Compared with the normal group,the contents of MMP-3,MMP-9,MMP-13,MMP-14 and TIMP-1 were not significantly different between the sham operation’s group group and normal group(P>0.05).At the end of the12 th week,MMP-3/TIMP-1 ratio in the control group was significantly higher than that in the control group,the sham operation’s group and the normal group,and the difference was statistically significant(P<0.05).There was no significant difference in the MMP-3/TIMP-1 ratio between the manipulation’s group and the sham operation’s group and the normal group(P>0.05).There was no significant difference in the ratio of MMP-3/TIMP-1 between the sham operation’s group and the normal group,which was not statistically significant(P=0.76).Conclusion: 1.Ligation of lower limb vein preparation method of IOH KOA model is desirable.2.Roubin manipulation can delay the degeneration of cartilage tissue.3.Roubin manipulation can significantly reduce the serum MMP-3,MMP-9,MMP-13,MMP-14 content of KOA model,increase TIMP-1 content,reduce the ratio of MMP-3/TIMP-1,might be one of the mechanism of roubin manipulation delay cartilage degeneration and the treatment of KOA.
