Objective To explore the mRNA expression of MMP3,MMP9,MMP13 and TIMP-1 in the development of adjuvant arthritis of rats (AA), especially their expression in cartilage and synovium of the metatarsophalanggales and knee articulations.Methods AA was yielded in male SD rats through subcutaneous injection of complete Freund's adjuvant(CFA) in their left hindpaws. Twenty-five SD rats were injected, amongst which five experimental rats were sacrificed 1, 2, 5, 7 and 9 weeks after injection respectively. The cartilages and their adjacent synovium of the articulations in affected metatarsophalanggales and their congruent-flank knees were obtained to detect by RT-PCR the expression of mRNA of MMP3,MMP9,MMP 13 and TIMP-1. Meanwhile, these carilages and synoviums were harvested to observe macropathologic changes during the ninth week.Results In the first two weeks, expression of the rats'MMP3 and MMP13 in paws'cartilage and synovium was detected, within which the rate of MMP3 was higher than that of MMP13. Furthermore, MMP3 lasted for five weeks while MMP13 for only two weeks. Meanwhile the expression of MMP9 was unstable; TIMP-1 was not detected until the seventh week. TIMP-1 could be located in rats'knee, but no expression of MMP3,MMP9 and MMP13 found in the whole course of this experiment. The synovium of the metatarsophalanggales articulations of the paw gave obvious inflammatory reactions in the ninth week, while the cartilages and synovium of the knee remained normal in this aspect.Conclusion It can thus suggest the balance among MMP 3, MMP 9, MMP 13 and...
