The Effect Of Hongjindan On TNF-α、IL-10 And IKK/NF-κBP65 Signaling Pathway Of Pelvic Inflammatory Disease Rats

Objective:Through the experiments with rats,to observe the therapeutic effect of Hongjindan Enema on rats of pelvic inflammatory disease model,to measure the concentration of TNF-a and IL-10 in serum of rats after treatment,the expression of uterine tissue of rats with TNF-a,and the protein expression of TNF-α,Ikk,IκB,NF-κBP65 in Rats’ uterine tissue,to explore the mechanism of Hongjindan’s treatment on pelvic inflammatory disease,to provide theoretical basis for clinical application.Method:To randomly divided 60 rats into six groups and each group with 10 rats,were normal group(sham operated group),model group,Positive control group(Fuke Qianjin Soft Capsule(FQSC)group),and Hongjindan in high,middle,low dose group(three groups).Excepted the normal group,the other groups were injected phenol mucilage to establish as rats model of pelvic inflammatory disease.After the model was successfully established,according to the experimental group,the sham operated group was treated with distilled water,the three groups of Hongjindan were treated with Hongjindan solution,and FQSC group was treated with FQSC.After 14 days of treatment,took of the serum and uterine tissue of rats.Observe the morphology of rats’ uterus through eyes and light morphology;the content of IL-10 and TNF-a in serum was detected by Elisa;the average optical density of TNF-α in uterine tissue was detected by IHC and the protein content of TNF-α,Ikk,IκB and NF-κBP65 were determined by western blotting.Result:1 The influence of morphologyThrough the observation by eyes and light morphology,we can see that the rat uterus had varying degrees of congestion and swelling,the pelvic adhesion degree of the model group was serious,adhere with the surrounding connective tissue and organ,and cannot be divided.The sham operated group was no adhesion and swelling.And each drug administration group both had improved,especially Hongjindan in high dose group and Positive control group.2 The influence of the concentration of TNF-a,IL-10 in rats’ serumThe level of TNF-a in model group was significantly higher than the sham operated group,which had a high degree of statistical significance(P<0.01).Among these,Hongjindan in high dose group and Positive control group were more close to the level of sham operated group,had no statistical significance with it(P>0.05).The level of IL-10 in model group was significantly lower than sham operated group,had a high degree of statistical significance(P<0.01).There were different degrees of increase after treated by all groups treated with medicine.Among these,Hongjindan in high dose group and Positive control group were more close to the level of sham operated group,had no statistical significance with it(P>0.05).3 The influence of the contents of TNF-α,Ikk,IκB and NF-κBP65 in uterine tissueThe average optical density of TNF-a in model group was significantly higher than all group else,had a high degree of statistical significance(P<0.01).Hongjindan in high dose group and Positive control group were more close to the level of sham operated group,had no statistical significance with it(P>0.05).The protein content of TNF-a,IKK and NF-κBP65 in model group was significantly higher than that in the sham operated group,had a high degree of statistical significance(P<0.01).There were different degrees of decrease,especially Hongjindan in high dose group and Positive control group were more close to the level of sham operated group,had no statistical significance with it(P>0.05).The protein content of IκB in model group was lower than that in the sham operated group,had statistical significance(P<0.05).There were different degrees of increase,especially Hongjindan in high dose group and Positive control group(P<0.01).Conclusion:Hongjindan could reduce the inflammatory cell infiltration of pelvic inflammatory disease model rats,could reduce congestion and swelling of the inflammatory tissue,could reduce the adhesion of pelvic inflammatory tissue with surrounding tissue.The mechanism of Hongjindan’s treatment on pelvic inflammatory disease maybe:(1)by increasing the expression of IL-10,to inhibit the inflammatory corpuscle reaction,to reduce the synthesis and activity of TNF-a and other pro-inflammatory factors;(2)by decreasing the expression of TNF-α,to reduce inflammatory cells,to reduce the inflammatory response,to accelerate the inflammation injury repair,and therefore to promote the body to return to normal;(3)by inhibiting the signal pathway of Ikk/NF-κBP65,to inhibit the activity of NF-κB,or by reducing the activation of NF-κB,to decrease the release of TNF-α and other inflammatory factors,thereby restoring the body’s immune balance.
