Objective: To observe the effect of BAISHAOQIWU granules on NOXs-OS-P38 MAPK inflammatory signaling pathways in the colon tissue in trinitrobenzen suiphonic acid(TNBS)-induced ulcerative colitis(UC)rats,and to investigate the underlying mechanisms.Method:72 male SD rats were divide into normal group,Model group1-5,totally 6 groups randomly,12 rats in each group.rats in Model groups were built as UC mode by trinitrobenzen suiphonic acid(TNBS).And2 days later,2 rats in each group were chosen randomly and they were narcotized to ensure the UC model was successfully built.Then,the rest rats were randomly divided into 6 groups(model group,SASP group and BAISHAOQIWU granules low,medium and high dose group,with normal group).Each group of rats were given intragastic administration with Corresponding drugs.Once a day,14 days continuously.After the Period of treatment,all mice were given a mark on their disease activity index(DAI)and colonic mucosa damage index(CMDI).Then the Colon tissue of mice were chosen to test the expression of P38 MAPK m RNA and interleukins-1β m RNA by HE staining and Fluorescence quantitative RT-PCR,and serum of rats were taken out to detect the expression of NOXs,ROS,P38 MAPK and IL-1β by Elisa.Results:(1)On DAI,CMDI and the microscopic pathological grading,the score of rats in treatment group was lower than model groups obviously(P< 0.01),all showed that BAISHAOQIWU granules can improve clinical symptoms of UC rats and promote healing of mucous membrane,compared with SASP group,the effect was fairly or better(P < 0.05).(2)Compared to the normal group,the expression of P38 MAPK m RNA,IL-1β m RNA in Colon tissue and NOXs,ROS,P38 MAPK and IL-1β in serum of rats in model groupwere rising(P < 0.01).(3)Compared to model group,the expression of P38 MAPK m RNA,IL-1β m RNA in Colon tissue and NOXs,ROS,P38-MAPK and IL-1β in serum of rats in the three BAISHAOQIWU granules groups decreased(P< 0.01);(4)Compared to SASP group,except for the expression of P38 MAPK m RNA in BAISHAOQIWU granules high dose group,had no significant difference(P > 0.05),other groups of indicators had significant difference(P< 0.01).Conclusion:BAISHAOQIWU granules can reduce the expression of proinflammatory cytokines IL-1β to treating UC rats by regulate NOXs-ROS-P38 MAPK signal pathway.
