Objective:To observe the molecular mechanisms of BAISHAOQIWU granules on treating Ulcerative colitis(UC).Method:60 rats with half males and half females were divide into normal group 、 Model group1-5,totally 6 groups randomly,10 rats in each group. Rats in Model groups were built as UC mode by trinitrobenzen suiphonic acid(TNBS).And 2 days later,2 rats in each group were chosen randomly and they were narcotized to ensure the UC model was successfully built.Then,the rest 40 rats in model groups were randomly divided into Blank model group, Positive control group and BAISHAOQIWU granules low、medium and high dose group,with normal group,totally 6 groups, 8 rats in one group. Each group of rats were given gavage treatment with Corresponding drugs.Once a day, 14 days continuously.After the Period of treatment,all rats were given a mark on their disease activity index(DAI)and colonic mucosa damage index(CMDI).Then the Colon tissue of rats were chosen to test the expression of NLRP3 m RNA and interleukins-1β m RNA by HE staining and Fluorescence quantitative RT-PCR,and serum of rats was taken out to detect the expression of NLRP3、Caspase-1and IL-1β by Elisa.Results:(1)On DAI and CMDI,the score of rats in blank model group was higher than rats in other groups obviously(P<0.01),and the DAI and CMDI score of rats in other groups had no significant differences(P > 0.05).(2)Compared to the normal group,the expression of NLRP3 m RNA、IL-1β m RNA in Colon tissue and NLRP3、Caspase-1、IL-1β in serum of rats in blank model group were rising(P<0.01).(3) Compared to blank model group,the expression of NLRP3 m RNA、IL-1β m RNA in Colon tissue and NLRP3、Caspase-1、IL-1β in serum of rats in the three BAISHAOQIWU granules groups decreased(P<0.01); And in the three groups,the degree of indicators of rats in high dose BAISHAOQIWU granules group fell more remarkable(P <0.01).Conclusion:BAISHAOQIWU granules can reduce the expression of proinflammatory cytokines IL-1β to cure UC by regulate NLRP3 inflammasome signal pathway.
