Objective: To investigate the changes of 16 species associated with intestinal diseases in Ulcerative clitis(UC)rats by real-time quantitative quantitative polymerase chain reaction(q PCR).To investigate the effect of experence Chinese medcine Anchang decoction on the changes of intestinal flora in UC rats and to explore the change character of intestinal flora in UC rats.To explore the mechanism of the treatment of UC and the clinical application of Anchang decoction promote the provision of theoretical basis,but also to deepen understanding of UC disease and a comprehensive understanding of UC intestinal flora changes provide the basis.Methods:(1)experimental group: 112 healthy rats were randomly divided into UC model group,normal control group,Mesalazine group,Bifidobiogen group,Anchang decoction low-,middle-and high-dose group,each group of16 rats.(2)experimental modeling: 2,4,6-trinitrobenzene sulfonic acid(TNBS)/acetic acid modeling method for modeling except the normal control group,the rats in each group were treated by intragastric administration on the 10 th day after modeling,and rats were sacrificed 7 days after intragastric administration.(3)subjects: rats in each group and their feces and colon tissues.(4)experimental method and test content: during the experiment,the activities of the rats were investigated,and the fecal traits and the occult blood were observed.The disease activity index(DAI)score was taken.Pathological analysis was performed to determine the mucosal lesion of the rats by the colon macroscopic damage index(CMDI).The Geboes index was used to evaluate the histopathological status of the colonic mucosa.The contents of the colonic contents were extracted from the colonic contents by SYBRGreen fluorescence.The results showed that the extracts were quantitatively determined by Delta-delta Ct and quantified by the quantitative method.Results:(1)the symptoms of the disease: The symptoms of the disease in UC model group were significantly and the DAI score was higher than the normal control group(p<0.05).The symptoms of the disease in the treatment group(Mesalazine group,Bifidobiogen group,Anchang decoction low-,middle-and high-dose group)appear to ease after intragastric administration and the DAI score was lower than that of the UC model group(p<0.05).The middle dose group of Anchang decoction was lower than that of the high dose group and the low dose group(p<0.05).The DAI score of the treatment group in 7th day was lower than that of the 4th day(p<0.05).The DAI score of the middle dose group of Anchang decoction group was lower than that of the Mesalazine group,Bifidobiogen group group on the 7th day(p<0.05).(2)the colonic tissue status of the rats in each group: About the CMDI score and the Geboes index,UC model group was higher than that in the normal control group(p<0.05),and the treatment group was lower than the UC model group(p<0.05),The middle dose of Anchang decoction group was lower than that in the high and low dose groups(p<0.05).(3)changes of intestinal flora in UC model group: compared with the normal control group,the relative expression of UC model group showed decreased in the Lactobacillus,Bifidobacterium,Clostridium,Bacteroides fragilis,Bacteroides(p<0.05),and in the Bacteroides,Bacteroides,Escherichia coli,Bacillus thuringiensis,Enterobacteriaceae,Enterococcus faecalis,Enterococcus faecalis showed increased(p<0.05).(4)changes of intestinal microflora in the Mesalazine group and Bifidobiogen group: compared with UC model group,the relative expression of Mesalazine group and Bifidobiogen group showed increased in Lactobacillus,Bifidobacterium,Clostridium,Bacteroides fragilis,Bacteroides multiforme,Bacteroides ovum,Bacteroides(p<0.05),while the expression level was lower than that of the normal control group(p<0.05);in the Bacteroides simplex the expression level of the Mesalazine group was higher than that of the UC model group(p>0.05),The expression level of Bifidobiogen group was higher than that of UC model group(p<0.05),at the same time,the two groups were lower than the normal control group(p<0.05);in the genus Fusarium,Botox,Bacteroides,Bacteroides,Enterococcus faecalis,Enterococcus faecalis and Escherichia coli,the relative expression levels of Mesalazine group and Bifidobiogen group were lower than that of UC model group(p<0.05),while lower than the normal control group(p<0.05);in the genus Bacteroides,the Mesalazine group was lower than the UC model group(p>0.05),higher than the normal control group(p<0.05),the Bifidobiogen group was lower than the UC model group(p<0.05),higher than the normal control group(p<0.05).(5)the changes of intestinal flora in Anchang decoction treatment group(Anchang decoction low-,middle-and high-dose group): compared with UC model group,The relative expression levels of the treatment group was rise in Lactobacillus,Bifidobacterium,Clostridium,Bacteroides fragilis,Bacteroides mult(p<0.05),and showed reduce compared with normal control group(p<0.05);In the treatment group,the expression level of Escherichia coli,Enterobacteriaceae,Enterococcus faecalis,Escherichia coli and Escherichia coli was lower than that in the UC model group(p<0.05),while higher than that of the normal control group(p<0.05);The low dose of Anchang decoction in Bacteroides simplex was higher than that in UC model group(p>0.05),while lower than that of the normal control group(p<0.05);The middle dose was higher than that of the UC model group(p<0.05),while lower than that of the normal control group(p<0.05);The high dose was higher than that of the UC model and normal control group(p<0.05).(6)Comparison of intestinal microflora changes in Anchang decoction treatment group: The middle dose of Anchang decoction Lactobacillus,Bifidobacterium,Fusobacterium,Bacteroides fragilis,Bacteroides bacillus,Bacteroides multiforme,Botox,Bacteroides bacillus,Enterobacteriaceae,Enterococcus faecalis,E.coli was closer to the blank control group than the other two groups(p<0.05);The high dose group was more close to the other two groups in the genus Bacteroides,Clostridium,O.coli and Gibbidium(p<0.05).(7)Comparison of intestinal microflora changes between Anchang middle decoction group and Mesalazine group、Bifidobiogen group:The relative expression level of the middle dose of Anchang Decoction group was more close to the normal control group in Lactobacillus,Bacteroides fragilis and Bacteroides than Bifidobiogen group(p<0.05),more close to the normal control group in Bifidobacterium,Clostridium,Bacteroides bacillus,Botox than Mesalazine group(p<0.05),and in Escherichia coli,Enterococcus faecalis,Enterococcus faecalis,Dendrobacter,Bacteroides bacillus,Osteobacterium,Bacteroides multiforme and Bacteroides was closer to the normal control group compared to Mesalazine group and Bifidobiogen group(p<0.05).Conclusion:(1)In this study,UC rat model in the disease symptoms,colonic injury and pathological manifestations are consistent with the performance of UC disease,UC rat model was established more successful,Anchang decoction and Mesalazine group and Bifidobiogen group has a therapeutic or alleviating effect on UC rats.(2)The changes of intestinal flora in UC rats were significantly different and characteristic.This change may play a role in the pathogenesis of UC,and the changes of this group may be used as the diagnostic marker of UC.(3)Anchang decoction had obvious regulation effect on the whole intestinal microflora of UC rats and was consistent with the combination of Mesalazine and Bifidobiogen,and the effect of Anchang decoction on UC intestinal flora may be the mechanism of its treatment of UC;(4)Compared with the high dose and low dose of Anchang decoction,the middle dose had a more significant effect on the symptoms and intestinal microflora of UC rats.At the same time,the regulation of Escherichia coli,Enterococcus faecalis,Enterococcus faecalis,Bacteroides bacillus,Bacteroides gibberella,Oocarbophilus,Bacteroides multiforme and Bacteroides was superior to that of Mesalazine and Bifidobiogen,indicating that From the relieve symptoms and intestinal flora regulationpoint of view,it is possible to improve the current clinical application dose.
