Objective:On the basis of the previous research of the research group,by observing the regulating effect of Anchangyuyang Decoction combined with Mesalazine on the intestinal flora of patients with ulcerative colitis,to explore its mechanism of treatment of ulcerative colitis.Methods:A total of 23 patients who met the UC inclusion criteria in the outpatient and inpatient departments of the Spleen and Gastroenterology Department of the Affiliated Hospital of Shandong University of Traditional Chinese Medicine from September 2017 to December 2020 were included as the observation group,and 10 healthy patients were selected as the control group.The observation group was given Anchangyuyang Decoction combined with Mesalazine orally for 12 weeks,while the control group received no treatment.Comparison of endoscopic Baron score and histopathological Geboes score of observation group before and after treatment;comparison of fecal flora difference between observation group and control group before and after treatment;comparison of fecal flora difference of observation group before and after treatment.Results:(1)Endoscopic Baron score:The observation group was significantly lower after treatment than before treatment(P<0.01).(2)Histopathology Geboes score:The observation group was significantly lower after treatment than before treatment(P<0.01).(3)Analysis of flora Alpha diversity:Shannon index and ACE index of the observation group were significantly lower than those of the control group before treatment(P<0.01),and Simpson index increased(P<0.01);after treatment,the observation group was compared with Shannon index and ACE before treatment.The index increased in varying degrees(P<0.01,P<0.05),and the Simpson index decreased(P>0.05).(4)Beta diversity analysis of bacterial flora:the clusters of samples collected in the observation group before treatment are far from the control group,and the clustering of samples in the group is poor;the clusters of samples collected in the observation group after treatment are closer to the control group,and the clusters of samples in the group are better.(5)Changes in the abundance of phylum horizontal flora:The abundance of Firmicutes in the observation group before treatment was lower than that in the control group,and the abundances of Actinobacteria,Proteobacteria,and Bacteroides were all increased;the observation group was thicker than before treatment The abundance of Bacteria increased,and the abundances of Actinobacteria,Proteobacteria,and Bacteroides all decreased.(6)Changes in the abundance of the genus horizontal flora:The observation group had lower abundances of Faeculus,Lauterella,and Microbacteria before treatment,and EscherichiaShigella,Streptococcus,and Bacteroides The abundance of,Corinthella,Haemophilus,and Enterococcus increased;after treatment,the observation group had increased abundance of Faeculus and Lauterella,and Escherichia-Shigella The abundance of spp,Streptococcus,Collinsella,and Haemophilus decreased,while the abundance of Bacteroides and Enterococcus did not change much.Conclusion:There are significant differences in the composition of fecal flora between UC patients and healthy people.The diversity and richness of fecal flora of UC patients decreases,the abundance of beneficial bacteria decreases,and the abundance of pathogenic bacteria increases.This is consistent with the results of domestic and foreign scholars.Anchangyuyang Decoction combined with Mesalazine not only has a significant effect in achieving UC endoscopic and histopathological relief and promoting deep mucosal healing,but also can regulate the fecal flora of patients with UC,increase its diversity,richness,and increase intestines Path beneficial bacteria,reduce pathogenic bacteria.Based on the results of the previous research of this research group,Anchangyuyang Decoction combined with Mesalazine can restore the balance of intestinal flora,affect the NF-κB and JAK/STAT signaling pathways to regulate inflammatory factors,and achieve the purpose of treating UC. |
