Objective:To observe the expression of Hrh1 and filaggrin gene on acute eczema rat’s back, to explore the possible mechanism of acute eczema treated by two different purslane extract effective components.Method: 50 SD rats were randomly divided into normal control group, model group, positive drug control group, purslane alcohol extract group, and purslane freshly squeezed fluid group. Except the normal group, the hair of rat’s abdomen were shaved by electric electric shaver, with an area of about 1 cm×2 cm. Then,2,4-dinitrochlorobenzene was used to make rats acur first sensitization. After 3 days,2,4-dinitrochlorobenzene was used again on the right back of rat, the area is 2 cm×2cm, the left back as the control site. To use excitation on rat every 3 days,3 times one day. Ninth days after treatment, the right back of the rat skin was marked erythema, edema, rash, erosion or exudation, tips to make a successful model. To stimulate the 10th day after administration, twice a day, each 1ml, nine consecutive days, finally rats were decapitated in the 21st day after the first sensitization, then clipped molding area skin which were given RT-PCR detection in each experimental group rats skin filaggrin gene expression, and immune group of detection of Hrh1 mean optical density.Results:First, compared with the normal group, model group rats skin tissue’s Hrhl protein content significantly increased, with significant statistical significance (P <0.01), and compared with the model group, Hrhl protein content of freshly squeezed rats skin tissue significantly decreased, (P< 0.01), tissue Hrhl protein content of positive control group and alcohol provided group rat skin decreased with very significant statistical significance, with statistical significance (P<0.05). Second, compared with the normal group, the 2-△△CT value of model group’s filaggrin gene was significantly smaller (P< 0.01), compared with the model group, the 2-△△CT value of portulaca oleracea freshly squeezed liquid group and positive drug control group’s filaggrin gene in rats skin tissue cells was significantly increased (P<0.01), the value of alcohol provided group was significantly increased (P< 0.05).Conclusion:The two purslane extracts can both constrict the blood vessels, inhibit capillary permeability and inflammatory swelling, so as to reduce the local eczema itching, edema symptoms. Purslane freshly squeezed liquid is better. One of the possible mechanisms is that the two purslane extracts can significantly reduce the level of rat skin itching factor Hrhl, obviously control rats’s scratched catch reaction and improve the skin lesions, the two purslane extracts also raised the expression of filaggrin gene in the eczema animal skin barrier system and accelerated filaggrin hydrolyzing into a variety of natural moisturizing factors which can effectively improve and repair the skin barrier function of rats, enhance the immune function of the body, thus playing a role in the treatment of acute eczema. |
