[Objective] Research group screening key genes of EB virus induced lymphoma in preliminary experiment: TOP2 A, HIST2H2 BE and FOS. This paper detect the protein expression of TOP2 A, HIST2H2 BE and FOS in tissue samples of malignant lymphoma and lymph node reactive hyperplasia, study the role and significance in lymphoma.[Materials and Methods] Collect paraffin tissue samples of 100 cases of malignant lymphoma and 40 cases of lymph node reactive hyperplasia from xiangya hospital, central south university. Classify according to lymphoma histopathological and immunohistochemical, there are 24 cases of Hodgkin’s lymphoma, 76 cases of non-hodgkin’s lymphoma. Apply immunohistochemical SP staining to detect TOP2 A, HIST2H2 BE, FOS protein expression in the tissue of lymphoma and lymph node reactive hyperplasia, apply in situ hybridization to detect expression of EBER in the tissue of lymphoma. Analyse the relationship between the expression of TOP2 A, HIST2H2 BE, FOS and its clinical pathological parameters.[Results]1. TOP2 A protein expression in the lymphoma and lymph node reactive hyperplasia and its relationship with clinical pathological parameters TOP2 A positive staining was located in the nucleus. The positive expression rate of TOP2 A protein in 100 cases of lymphoma tissue was 88%, including Hodgkin, B cells, T cell lymphoma was 83.3%, 90% and 88.5% respectively. In lymph node reactive hyperplasia, the positive expression rate of TOP2 A protein was 90.4%. Compared to lymphoma and lymph node reactive hyperplasia, there was no significant difference of TOP2 A protein positive expression rate(p > 0.05). Compared to Hodgkin’s and non-hodgkin’s lymphoma, there was no significant difference of TOP2 A protein positive expression rate(p > 0.05). Compared to B cell and T cell lymphoma, there was no significant difference of TOP2 A protein positive expression rate(p > 0.05). TOP2 A protein positive expression had no correlation with patients’ age, position(p > 0.05), had correlation with patients’ gender(p < 0.01). 2. HIST2H2 BE protein expression in the lymphoma and lymph node reactive hyperplasia and its relationship with clinical pathological parameters HIST2H2 BE positive staining was located in the nucleus. The positive expression rate of HIST2H2 BE protein in 100 cases of lymphoma tissue was 85%, including Hodgkin, B cells, T cell lymphoma was 91.7%, 80% and 88.5% respectively. In lymph node reactive hyperplasia, the positive expression rate of HIST2H2 BE protein was 42.5%. Compared to lymphoma and lymph node reactive hyperplasia, there was significant difference of HIST2H2 BE protein positive expression rate(p < 0.01). Compared to Hodgkin’s and non-hodgkin’s lymphoma, there was no significant difference of HIST2H2 BE protein positive expression rate(p > 0.05). Compared to B cell and T cell lymphoma, there was no significant difference of TOP2 A protein positive expression rate(p > 0.05). HIST2H2 BE protein positive expression had correlation with patients’ gender, age, position(p < 0.01). 3. FOS protein expression in the lymphoma and lymph node reactive hyperplasia and its relationship with clinical pathological parameters FOS positive staining was located in the nucleus. The positive expression rate of FOS protein in 100 cases of lymphoma tissue was 51%, including Hodgkin, B cells, T cell lymphoma was 54.2%, 44% and 61.5% respectively. In lymph node reactive hyperplasia, the positive expression rate of FOS protein was 95%. Compared to lymphoma and lymph node reactive hyperplasia, there was significant difference of FOS protein positive expression rate(p < 0.01). Compared to Hodgkin’s and non-hodgkin’s lymphoma, there was no significant difference of FOS protein positive expression rate(p > 0.05). Compared to B cell and T cell lymphoma, there was no significant difference of TOP2 A protein positive expression rate(p > 0.05). FOS protein positive expression had no correlation with(p > 0.05), had correlation with patients’ age, gender, position(p < 0.01). 4. EBER detection in the lymphoma and its relationship with clinical pathological parameters EBER positive staining was located in the nucleus. The positive rate of EBER in 100 cases of lymphoma tissue was 26%, including Hodgkin, B cells, T cell lymphoma was 41.7%, 8% and 46.1% respectively. Compared to Hodgkin’s and non-hodgkin’s lymphoma, there was significant difference of EBER positive rate(p < 0.05). Compared to B cell and T cell lymphoma, there was significant difference of EBER positive rate(p < 0.01). EBER positive rate had correlation with types of lymphoma, patients’ gender, age, position(p < 0.01).[Conclusions] 1. TOP2 A in lymphoma and reactive hyperplastic lymphoid tissue all had a high positive rate, there was no significant difference. 2. HIST2H2 BE protein expression was up-regulated in lymphoma tissue than in lymph node reactive hyperplasia. 3. FOS protein expression was down-regulated in lymphoma tissue than in lymph node reactive hyperplasia.
