| Objective: Studies in Xiaoer Qingfei Zhike oral liquid in expression of matrix metalloproteinase-9(MMP-9)and matrix metalloproteinase inhibitor-1(TIMP-1)in mice lung tissue to prove that Xiaoer Qingfei Zhike oral liquid treatment of acute bronchitis effect model mice,further study in children with acute bronchitis Qingfeizhike oral mechanism of action,provide objective laboratory basis for its clinical efficacy.Methods: 80 healthy 4-week-old Kunming mice were randomly divided into model group 64,control group 16.After a mouse model of acute bronchitis smoked were successfully prepared,and then the mice were randomly divided into five groups:model group,pediatric oral Qingfeizhike high,medium and low dose group,positive oral pediatric cough heat the control group,and were administered with corresponding drugs treatment.After 1 week treatment,taking immunohistochemistry to test the content of MMP-9 and TIMP-1 in mice lung tissue.Results: 1 MMP-9 assay results: Xiaoer Qingfei Zhike oral liquid high,medium and low dose group,Xiaoer Qingrezhike oral liquid positive control group and model group lung tissue content of MMP-9 were higher than the normal group,and compared with the normal group,there were different(P <0.05);High,medium and low dose group and the control group content of lung tissue MMP-9 were lower than the model group,and were different(P <0.05)compared with the model group;Content control mice lung tissue MMP-9 are high-dose group compared with three concentrations and were different(P <0.05)compared with treatment groups;Xiaoer Qingfei Zhike oral liquid high,there was no difference(P> 0.05)between medium and low dose groups.2 TIMP-1 assay results:Model group and the control group of mice lung tissue content of TIMP-1 were lower than the normal group,and compared with the normal group,there were different(P <0.05);High,medium and low dose group and the control group content of lung tissue TIMP-1 was higher than the model group,and were different(P <0.05)compared with the model group;High,medium and low dose group content of TIMP-1 in lung tissue were higher,and were different(P <0.05)compared with the control group;Xiaoer Qingfei Zhike oral liquid high,there was no significant difference(P> 0.05)compared to between medium and low dose groups.Conclusion: 1 Compared with the normal group,the content of TIMP-1 was lower,and the content of MMP-9 was higher in the lung tissue of acute bronchitis model mice,this shows that MMP-9,TIMP-1 participates and back-adjusts the formation of acute bronchitis in mice;2 Xiaoer Qingfei Zhike oral liquid can significantly increase the expression of TIMP-1 in mice lung tissue,reduces the expression of MMP-9,and can regulate the expression of MMP-9 and TIMP-1.Its efficacy is superior Xiaoer Qingrezhike oral positive control group,and for the clinical curative effect of Xiaoer Qingfei Zhike oral liquid in treatment of acute bronchitis provide a more adequate laboratory evidence. |
