Objective:To prepare the mice model that can simulate acute bronchitis of pediatricbetter.Using modern molecular biology techniques to determine the levels about the tissueof mice lung transcription factor GATA-3and T-bet.Analysis the expression of Xiao’erQingfei Zhike oral liquids about the tissue of mice lung transcription factor GATA-3andT-bet.Then we verify the curative effect of Xiao’er Qingfei Zhike oral liquids on mice ofacute bronchitis model.And explore the mechanism of Xiao’er Qingfei Zhike oral liquids onacute bronchitis,in this way can we have strong laboratory evidence for clinical medication.Methods: Choosing80Kunming species mice which are heathy and randomly divided intonormal group with16and model group with64. We use smoked to prepared for acutebronchitis model.Every time we smoked them30minutes,2times a day (every morning andevening),for one week.After building the successful model group mice,we randomly dividedthem into5groups. One group is the model group, and the others are the Xiao’er QingreZhike oral liquid positive control treatment group,Xiao’er Qingfei zhike Oral Liquid thehigh,middle and low dose treatment group.which are respectively treated by and Xiao’erQingre Zhike oral liquids.Xiao’er Qingfei Zhike oral liquids with high, middle and lowdose,the model group treated by equal volume saline.After one week treatment,we take thelung tissues of all the mice by immunohistochemical method to text the integral opticaldensity of mouse lung tissue of transcription factor GATA-3and T-bet.Results:1Mice lung tissue of GATA-3immunohistochemical results:Every treatmentgroup and model group,compared with the normal group has a very significantdifference(P<0.01);Xiao’er Qingfei zhike Oral Liquid the high dose group,middle dosegroup,and low dose treatment group between the each two groups had no differences(P>0.05);Xiao’er Qingfei Zhike Oral Liquid high-dose group,middle dose group low dosetreatment group compared with the Xiao’er Qingre zhike oral positive control treatmentgroup showed a very significant difference(P<0.01);2T-bet immunohistochemical results of the lung tissue of mice: model group Xiao’er Qingre zhike oral positive control treatmentgroup and Xiao’er Qingfei zhike Oral Liquid the low dose treatment group compared to thenormal group had a very significant difference (P<0.01);both of Xiao’er Qingfei zhike OralLiquid the high dose treatment group and middle dose treatment group compared to thenormal group showed significant difference(P<0.05),Xiao’er Qingfe zhike Oral Liquid thehigh dose treatment group,middle dose treatment group,low dose treatment group betweeneach two groups had no differences (P>0.05)and compared with the Xiao’er Qingre zhikeoral positive control treatment group showed a very significant difference(P<0.01);Conclusion:1The GATA-3integral optical density about lung tissue of model group micehigher than normal group, and the T-bet integral optical density lower than normal group,prompt cytokines balance disorder in the model mice with acute bronchitis, and GATA-3and T-bet imbalance expression of is likely to be one of the important reasons for theformation of airway inflammation;2Xiao’er Qingfei Zhike oral liquids can inhabit the levelof the the lung tissue of GATA-3, increase the level of lung tissue of T-bet and correctingthe imbalance, then regulating Th1and Th2cells balance to inhibit airway inflammation,which provides a new way for the treatment of acute bronchitis;3Xiao’er Qingfei Zhikeoral liquids has better curative effect on acute bronchitis model mice, and its curative effectis obviously better than the control group.
