Objective: Anemia,as an important complication with children acute leukemia,which the mechanism of action is unclear.Recent studies showed that the incidence of childhood acute leukemia and prognostic significant differences between individuals.Mechanism of acute leukemia caused by anemia is more complex.It may be related to levels of inflammatory factors,erythropoietin obstacles,Epo generation,barriers of iron absorption,utilization of radiotherapy and chemotherapy and so on.In this study,through to analysis the expression and correlation of Hepcidin,IL-6,Epo,transferrin and ferritin of tissues with children acute leukemia.To explore the Hepcidin and anemia are characteristic relationships with children acute leukemia and provide theoretical support for the clinical treatment.Methods: The 30 cases of acute leukemia in children with newly diagnosed leukemia called acute untreated before therapies group,the group through chemotherapy achieved complete remission after 1-2 months called acute leukemia during remission group,30 healthy children as a control group.Determination of 30 cases of acute leukemia in children with before treatment group,during remission group and 30 healthy controls the expression of serum Hepcidin,IL-6 and Epo by enzyme-linked immunosorbent assay(ELISA).By testing with the anemia and iron associated molecular RBC,Hb,Plt,TRF and Ferritin.We are analysis of correlation between the index and the difference between the level of expression.To study the correlation between Hepcidin and anemia among the children acute leukemia.Results:1 The gender of acute leukemia group compared with control group had no significant difference(P=0.438)by χ~2 test in children.The age of acute leukemia group compared with control group was no significant difference(P=0.691)by independent sample t-test analysis.2 Through analysis anemia biochemical indices data of the before therapies,during remission and control group,three groups of subjects serum RBC,Hb,Plt expression level were significant difference(P<0.01)by analysis of variance(ANOVA).RBC,Hb expression level in three groups comparisons between groups were statistically different(P<0.01).The Plt expression level was statistically significant(P<0.01)between the before therapies and control groups,before therapies group and during remission group.But it was no significant difference(P=0.09)between the remission group and the control group.Three groups of subjects RBC,Hb and Plt level of expression were highest in the control group,followed by remission,the lowest initial treatment group.3 Childhood leukemia with before therapies group of Hepcidin,IL-6,Epo expression and ferritin levels were higher than during remission group and the control group,while the expression of transferrin before therapies group was lower than remission group and the control group.Three groups of subjects serum of Hepcidin,IL-6,Epo,TRF and ferritin concentration difference was statistically significant(P<0.01)by analysis of variance(ANOVA),between Hepcidin group show were differences in the groups were statistically significant(P<0.01);between IL-6 group show among all groups were statistically significant differences(P<0.05);among Epo group comparison showed early treatment group and leukemia remission group(P<0.01)and de novo leukemia group and the healthy control group(P<0.01)was statistically significant,but the difference between leukemia remission group and control group was not statistically different(P=0.066);between TRF group were differences among groups were statistically significance(P <0.05);between ferritin group of newly diagnosed patients and healthy controls leukemia remission group(P<0.01)and leukemia remission group and the healthy control group(P<0.01)with statistical significance,and early treatment leukemia remission group and the difference between groups was not statistically difference(P=0.956).4 Correlation between Hepcidin with other molecules show that Hepcidin and Hb(r=-0.414,P=0.023),Hepcidin and Plt(r=-0.479,P=0.007),Hepcidin and TRF(r=-0.418,P=0.021)were negatively correlated;Hepcidin and IL-6(r=0.709,P<0.001)expression of Hepcidin and Ferritin(r=0.583,P=0.001)were positively correlated;Hepcidin with EPO(r=0.055,P=0.774)no correlation in the before treatment of acute leukemia.Conclusions:1 Hepcidin expression levels is positive correlate with clinical symptoms of anemia with acute leukemia.2 Hepcidin,IL-6,Epo,and serum ferritin expression in acute leukemia was significantly increased.While after chemotherapy remission they were decreased significantly.TRF decreased expression in the serum of acute leukemia and increased expression in after chemotherapy remission.They were could be used as indicators of anemia in children with acute leukemia progression and treatment an important indicator.3 Hepcidin expression and serum IL-6,ferritin correlated with Hb,Plt,TRF negative correlation,while according to the expression of these changes and provide guidance for further study of the molecular mechanisms of Hepcidin with acute leukemia.
