Objective:To study the expression of erythropoietin receptor(EPOR) in leukemic cells and the relationship of serum erythropoietin and anemia in acute leukemia patients,to supply a new theoretical basis for the cytokines therapy in acute leukemia with anemia.Methods:Samples were collected from non-remission and recurrent acute leukemia patients(leukemic cells>50% with the exception of M6)in Shanxi province hospital and the second clinical medical college of Shanxi medical university from May 16,2005 to December 21,2005, diagnosis according to the WHO classification.Thirty acute leukemia samples were investigated:15 men and 15 women,age from 3 to 74,the average age is 31.2±19.0 years old, including 21 acute myelogenous leukemia and 9 acute lymphocytic leukemia. Total RNA of marrow leukemic cells was isolated with TRIzol reagent according to the manufacturer's protocol.The quantity of total extracted RNA samples were then examined using spectrophotometric A260 measurements and separated by 1% agarose gels electrophoresis. EPOR was detected in 30 AL patients by using reverse transcription polymerase chain reaction,the relatively uniform levels of ?-actin transcript expression between the various samples in this study allowed the use of ?-actin as the standard.After 30 cycles PCR products were separated by 1% agarose gels electrophoresis, and photographed with Imaging system, The relative level of expression of EPOR mRNA was therefore computed with respect to the mRNA expression level of the reference ?-actin transcript.The level of serum erythropoietin was detected by chemiluminescence analysis as well as hemoglobin by automatic blood counting instrument producted by Japan.Results:EPOR was expressed in 18 of 30 AL patients.The percentage of EPOR expression was 61.9%(13/21) in AML and 55.6%(5/9) in ALL,The percentage of EPOR expression was no significant difference between AML and ALL. The percentage of EPOR expression was 60% in AL and was significantly lowr than that of control group (86.7%)(P<0.05),the relative quantity of EPOR expression in AML(0.4236±0.4433) was higher than ALL(0.1737±0.1922)(P<0.05),the average quantity of EPOR expression in AL was 0.3486±0.3991,significantly lowr than that of control group(0.6526±0.3231)(P<0.01).The average level of sEPO in 30 AL patients was 322.3±232.0,significantly higher than control group(44.8±55.6)(P<0.01),and there was negative correlation between the level of sEPO and Hb(r=– 0.658,P<0.01).Conclusion:EPOR was expressed in leukemia cells,but the expressive level was low.The percentage of EPOR expression was no significant difference between AML and ALL.the mechanism of negative feedback to anemia in acute leukemia is intact,anemia of acute...
